    ABSTRACT Objective To explore the effect of mammalian target of rapamycin (mTOR) inhibitor everolimus on radiosensitivity of human non-small cell lung cancer cell line in vitro by using everolimus to inhibit mTOR signaling pathway of A549.Methods Human non-small cell lung cancer cell line A549 was subjected to radiation alone or in combination with everolimus treatment.The 50%inhibition concentration (IC50) of everolimus in A549 cells was detected by methylthiazol tetrazolium (MTT) assay in vitro.Everolimus at the 20%inhibition concentration (IC20) was used to pretreat A549 cells for 24 h.Cells were then irradiated by X-ray with 2,4,6,8 Gy.The cell survival fraction was computed by clone formation.Cell survival curve was fitted by multitarget one-hit model,and mean lethal dose (D0),dose quasithreshold (Dq),survival fraction at 2 Gy (SF2),and sensitization enhancement ratio (SER) were calculated.The expression of γ-H2AX was determined by Western blotting and then the relative gray values were analyzed.Results Everolimus significantly improved the sensitivity of A549 cells to radiation.The D0,Dq and SF2of everolimus + irradiation group were significantly lower than those of irradiation group.The SER was 1.36.The residual amount of γ-H2AX protein in the everolimus + irradiation group was significantly higher than that of the irradiation group.Conclusion Everolimus inhibiting mTOR signaling pathway can increase the radiosensitivity of A549 cells.

    ABSTRACT Objective To explore the protective mechanism of Dendrobium nobile lindl (DNL) decoction on the expression of peroxisome proliferator activated receptor gamma (PPARγ) in the renal cortex of diabetic nephropathy (DN) rats.Methods Sixty SD rats were randomly divided into six groups (n=10 per group) as follows: normal control group (NC),diabetic nephropathy control group (DN),rosiglitazone group (RGZ),the Dendrobium nobile lindl low (LD),medium (MD) and high (HD) dose groups.After DN rat model was established,the rats were administrated with respective medications for 12 weeks,respectively.The renal pathology of rats was observed.The mRNA and protein expression of PPARγ in the renal cortex were detected via Real-time PCR and Western blotting,respectively.Results High dose DNL decoction significantly alleviated thickening of kidney tissue basement membrane and fusion of foot process in model rats.The levels of PPARγ mRNA and protein expression in the MD and HD groups were significantly increased as compared with the DN group (P<0.05,P<0.01).Conclusion DNL decoction can effectively reduce kidney injury by up-regulating the PPARγ mRNA and protein expression in diabetic nephropathy rats.

    ABSTRACT Objective To observe the effect of gypenoside on lipid peroxidation and hepatic lesion in rats with type 2 diabetes mellitus and nonalcohol fatty liver disease.Methods Totally,65 SPF male SD rats were randomly divided into blank control group (group N),NAFLD model group (group NM),and NAFLD with T2DM model group.The NAFLD with T2DM model group was further divided into three subgroups: JH group,perfused with 1 g•kg-1•d-1 GPS;JL group,perfused with 0.5 g•kg-1•d-1 GPS;model control group,perfused with the same volume of water.Blood sugar,triglycerides (TG),total cholesterol (TC),alanine aminotransferase (ALT),aspart aminotransferase (AST),adeponectin (ADP) in the plasma were measured.TG,malondialdehyde (MDA),and superoxide dismutase (SOD) in the liver tissue were also tested.Results ADP level was (7.46±1.12),(3.58±0.98),(4.89±1.02),(4.79±1.01) and (4.13±0.89) ng•mL-1 in N,M,NM,JH and JL groups,respectively.The ADP level was significantly higher in group JH and JL than in group M (P<0.01),and significantly higher in group JH than in group JL (P<0.05).MDA level was (2.98±0.09),(4.22±0.11),(3.66±0.10),(3.72±0.11),(3.99±0.13) nmol•mL -1 in N,M,NM,JH and JL groups,respectively.The MDA level was significantly lower in group JH and JL than in group M (P<0.01),and significantly lower in group JH than in group JL (P<0.05).SOD level was (240.8±17.4),(149.9±20.6),(181.6±19.4),(209.8±19.2),(189.4±18.9) U•mL-1in N,M,NM,JH,and JL groups,respectively.SOD level was significantly higher in group JH and JL than in group M (P<0.01),and significantly higher in group JH than in group JL (P<0.05).TG level was (28.98±1.68),(214.46±5.44),(198.46±6.98),(142.87±6.64) and (164.92±7.56) mg•g-1 in N,M,NM,JH and JL groups,respectively.TG level was significantly lower in group JH and JL than in group M (P<0.01),and significantly lower in group JH than in group JL (P<0.05).ALT and AST were significantly lower in group JH and JL than in group M (P<0.01),and significantly lower in group JH than in group JL (P<0.05).Conclusion The lipid peroxidation in the liver of rats with T2DM complicated with NAFLD can be reduced by gypenoside,and hepatic lesion may be alleviated through inhibition of lipid peroxidation.

    ABSTRACT Objective To demonstrate the mechanisms underlying the efficacy of telmisartan in diabetic nephropathy,and discuss the role of PPARγ in this process.Methods The diabetic nephropathy rat models were established and randomly assigned to control group,telmisartan group (5 mg•kg-1•d-1) and combination of telmisartan and PPARγ inhibitor group (telmisartan: 5 mg•kg-1•d-1;GW9662: 0.5 mg•kg-1•d-1).After 12 weeks of treatment,the biochemical indexes of blood and urine,kidney weight,renal pathology in each group of diabetic nephropathy rats were measured and compared.The levels of IL-1,IL-6 and TNF-α in blood of each group were detected by ELISA and compared.The levels of HGF and activated NF-κB (p65) in renal tissue of each group were detected by Western blotting and compared.Results In diabetic nephropathy rats,telmisartan lowered the levels of serum glucose,serum creatinine,urinary protein and kidney weight,decreased the glomerular volume,mesangial proliferation and inflammatory cell infiltration,reduced blood levels of IL-1,IL-6,and TNF-α,and decreased level of activated NF-κB (p65) in renal tissue.The level of HGF in renal tissue was elevated by telmisartan.Nevertheless,these changes were partly reversed by PPARγ inhibitor GW9662.Conclusion PPARγ presents an important role in treatment of diabetic nephropathy by telmisartan.

    ABSTRACT Objective To investigate the potential mechanism and protective effects of osthole on rats with focal cerebral ischemia-reperfusion injury.Methods Middle cerebral artery occlusion was induced by the suture-occluded method.After 2 h of cerebral ischemia and 24 h of reperfusion,scores of neurological deficits and infarct volume were evaluated.The levels of SOD,MDA,GSH and ATPase were also determined.Results Scores of neurological deficits and infarct volume were lower in the osthole groups than in the model group.Meanwhile,the activity of SOD and GSH content were increased,while the MDA content was decreased in osthole groups.Conclusion Osthole exerts protective effects on rats with focal cerebral ischemia-reperfusion injury.

    ABSTRACT Objective To explore the therapeutic effect of cyclovirobuxine-D (CVB-D) on heart failure by observing the influences of CVB-D on cardiac function,myocardial morphology and structure in rats with heart failure.Methods The rat model of heart failure was established by ligating left anterior descending coronary artery,and then the rats were randomly divided into 5 groups,plus the sham group.Echocardiography was used to assess cardiac function,including EF,LVSF,SV,HR,CO and so on.Myocardial tissue section was obtained.Change in the myocardial tissue was observed by HE staining.Myocardial ultra-structure was observed by using transmission electron microscope.Results As compared with the model group,EF and LVSF were significantly increased in medium and high dose CVB-D groups (P<0.05 or P<0.01),SV was increased in low and medium dose CVB-D groups (P<0.05),CO was increased in low,medium and high dose CVB-D groups (P<0.05),LVIDs was decreased and LVPWs was increased in high dose CVB-D group (P<0.05).Under light microscope,changes of myocardial structures were observed in different CVB-D dose groups: myocardial fiber gradually became tidy,stripes became clear,and contour became clear.The changes of myocardial ultra-structure were observed under transmission electron microscope: the myocardial fiber breakage was reduced and the number of mitochondria was increased,swelling degeneration was alleviated.Conclusion CVB-D significantly improved cardiac contraction and blood pumping function of rats with heart failure,exerted mitigative effect on damage of cardiac muscle fiber and mitochondria.It suggests that CVB-D may be effective in the treatment of heart failure.

    ABSTRACT Objective To investigate the effect of emodin on hypertrophic scar fibroblasts (HSFs) and explore the underlying mechanism.Methods HSFs were treated by emodin at final concentrations of 0,20,40,and 80 μmol·L-1,respectively,in the cultural media.Forty-eight hours later,the cells were subjected to MTS assay and flow cytometry assay with annexin V and propidium iodide as dyeing indicators.Whole cell lysates from the cells of every group were subjected to Western blotting to measure the protein expression levels of ERK1/2,Bcl-2,Mcl-1 and RIP1.Results The cell viability of HSFs was inhibited by emodin in a dose dependent manner.The mortality rate of HSFs treated with emodin for 48 h at the concentrations of 40 and 80 μmol·L-1 were 28.6%and 68.0%,respectively,which was significantly higher than that of the control group (P<0.01).Pretreatment with Z-VAD-FMK could partially reduce the mortality caused by emodin (P<0.05).Phosphorylation of ERK1/2 and the expression of RIP1 and Mcl-1 were inhibited by emodin.Conclusion Down regulation of ERK1/2,RIP1 and Mcl-1 by emodin may account for the inhibited proliferation and increased cell death of HSFs.

    ABSTRACT Objective To observe the effect of recombinant human granulocyte/macrophage colony stimulating factor (rhGM-CSF) and nano-silver as treatment for skin with deep Ⅱ degree burn.Methods Deep Ⅱ degree burn Wistar rat model was established.The rats were randomly divided into three groups,petrolatum treatment group (group A,n=30),nano-silver treatment group (group B,n=30),and rhGM-CSF treatment group (group C,n=30).The pathological changes of wound of the three groups were observed 1,4,7,10,14 and 21 days after the treatment.The concentration of VEGF in serums was measured with ELISA.The levels of HIF-1α mRNA expression were detected by RT-PCR.Results On day 10,neovascularization developed in groups A,B and C.Healing rate of the wound was the highest in group C,lowest in group A,with significant differences among the three groups on day 14 and day 21 (P<0.05).VEGF level peaked on day 14 in group A,and on day 10 in groups B and C.There were significant differences in VEGF level on day 1 between group A and groups B,C,on day 7 between group A and group C,on day 10,14 and 21 among the three groups (P<0.05),but no significant differences on day 4 among the three groups.Conclusion rhGM-CSF and nano-silver treatment can accelerate neovascularization of wound,and rhGM-CSF is better than nano-silver.

    ABSTRACT Objective To observe the clinical efficacy and safety of yiqing capsule in the treatment of acne with concomitant constipation.Methods A randomized controlled method was used.One hundred and forty-five patients with acne and concomitant constipation were randomized into treatment group (n=75) and control group (n=70).Patients in the treatment group received yiqing capsule orally and compound tretinoin gel topically,while patients in the control group received topical compound tretinoin gel only.Both groups were treated for 3 weeks.Results The therapeutic effect of the treatment group was much better than that of control group (P<0.01).No obvious adverse reactions were observed in either group.In the treatment group,77.4%of patients were relieved from constipation,the ratio being much higher than the control group (P<0.01).The chance of acne relief was remarkably higher in patients whose constipation were relieved (P<0.01).The Spearman correlation index between acne relief and constipation improvement was 0.699 (P<0.01).Conclusion Combination therapy of yiqing capsule and compound tretinoin gel is effective in the treatment of acne with no significant adverse reactions.The constipation improvement from yiqing capsule can contribute to the efficacy towards acne.

    ABSTRACT Objective To measure the renal cortex blood flow level in patients with stage 3-4 chronic kidney disease,and to observe the effect of probucol on renal cortex blood flow and renal function.Methods Twenty patients with stage 3-4 chronic kidney disease in our hospital were randomly divided into treatment group (10 patients) and observation group (10 patients).Ten healthy subjects were chosen as the control group.Dynamic sonographic tissue perfusion measurement (DTPM) was used to evaluate the mean flow intensity (Imix),mean flow velocity (Vmix) and tissue resistance index (TRI) in the three groups.Patients in the observation group were treated with standard therapy;the treatment group was given probucol 500 mg,twice daily in addition to standard therapy for 6 months.The renal cortex blood flow level after treatment was measured in the treatment group and observation group.The serum BUN,SCr,TC,TG,LDL,HDL,hs-CRP,ox-LDL,PA,ALB before and after treatment were measured and compared between the treatment group and observation group.Results Imix and Vmix were significantly lower,and TRI was significantly higher in the treatment group and observation group than in the control group (P<0.01).After 6 months of treatment,Imix and Vmix in the treatment group were significantly increased and TRI significantly decreased as compared with the baseline and the control group (P<0.01).After treatment,Imix and Vmix were significantly decreased and TRI was significantly increased in the observation group (P<0.05).In the treatment group,the serum BUN,SCr,TC,TG,LDL,hs-CRP and ox-LDL decreased and PA,ALB and HDL increased after treatment as compared with the baseline and the observation group (P<0.05 or P<0.01).Conclusion The renal cortex blood flow level is significantly lower in patients with stage 3-4 chronic kidney disease than in the healthy people.Probucol can improve the renal cortex blood flow level significantly,protect the renal function,and improve microinflammatory state,nutrition status and dyslipidemia.

    ABSTRACT Objective To study the effect of dexmedetomidine hydrochloride injection (Dex) on awakening quality in pediatric patients with congenital heart disease undergoing fast-track anesthesia and whole video-assisted thoracoscopic surgery.Methods One hundred and twenty pediatric patients (ASA Ⅰ or Ⅱ level) with congenital heart diseases were randomly chosen for this study.They received femoral artery and vein cannulation to establish cardiopulmonary bypass and three small holes were obtained in the right chest for the purpose of undergoing the whole video-assisted thoracoscopic surgery on repairmen of atrial septal defect or simple ventricular septal defect.They were randomly divided into two groups: Dex-assisted fast-track anesthesia group (D group,n=60) and general fast-track anesthesia group (C group,n=60).Each one in group D was injected with loading dose of Dex (1 μg&#8226;kg-1),then was intraoperatively infused with maintenance dose of 0.5 μg&#8226;kg-1&#8226;h-1.Children in group C were given the same dose of saline in the same way.Hemodynamic changes in the two groups at each time point: before anesthesia (t0),at the time of extubation (t1),5 min post-extubation (t2),10 min post-extubation (t3),15 min post-extubation (t4),30 min post-extubation (t5) and at the time of transferring out of CICU (t6),respectively,were observed.Restlessness extent,incidence,time of eye opening after calling,fully awaking time,extubation time,the time of transferring out of CICU and VAS scores,were also recorded.Results From t0 to t2-t5,SBP in D group was decreased from (114.2±10.5) mmHg to (107.2±10.3) -(105.3±11.3) mmHg,DBP decreased from (61.3±9.2) mmHg to (58.8±7.8) -(57.3±6.3) mmHg,and HR gradually decreased from (95.2±15.7) time&#8226;min-1 to (85.7±13.7)-(83.3±12.6) time&#8226;min-1,with significant differences (P<0.05).Compared with C group,SBP and DBP at t2-t5 were decreased significantly (P<0.01),and HR at t1-t5 decreased significantly (P<0.01).Overall incidence of agitation was significantly higher in group C than in group D (48.3%vs.16.7%,P<0.01).Incidence of moderate and severe agitation were significantly higher in group C than in group D (18.3%vs.6.7%,and 20%vs.1.7%,respectively,P<0.01).In group D,time of eye opening after calling,fully awaking time,extubation time,and the time of transferring out of CICU were prolonged,without significant difference as compared with group C (P>0.05).VAS at 30 min after extubation was significantly higher in group C than in group D(4.7±0.7 vs.2.4±0.6,P<0.05).Conclusion Injection of Dex with loading dose (1 μg&#8226;kg-1) and intraoperative infusion of maintenance dose of 0.5 μg&#8226;kg-1&#8226;h-1 in children with congenital heart disease undergoing fast-track anesthesia and whole video-assisted thoracoscopic surgery could be conducive to maintain hemodynamic stability,reduce restlessness occurrence,enhance security during awakening process,and alleviate post-operative pain.

    ABSTRACT Objective To explore the efficacy of nutritional intervention in patients with chronic obstructive pulmonary disease (COPD) and malnutrition risks.Methods From Jan.,2008 to Dec.,2012,829 COPD patients with NRS2002 score ≥ 3 in Qinzhou Second People's Hospital were enrolled in this study.Patients were randomized into control group (254 cases) and treatment group (575 cases) by random numerical table of SPSS 13.0 statistic software.Patients without contraindication to enteral nutrition were given enteral nutrition support,while those with contraindication to enteral nutrition were given parenteral nutrition support.Patients in the treatment group received intensive support with fortified nutrition,whereas patients in the control group received routine nutrition treatment.All other treatment Methods were the same between the two groups.Telephone follow-up lasted for 3 years in both groups after discharge.Patients in the treatment group with NRS2002 score ≥ 3 were given guidance on nutrition food intake.No nutrition guide was given to the control group.Times of acute attack,times and duration of mechanical ventilation,mortality rate,and NRS2002 score three years after the treatment were compared between the two groups.Data were analyzed by multi-factor Logistic regression analysis to understand the nutritional factors of COPD patients affecting their mortality rate.Results After 3 years of follow-up,times of acute attack,times and duration of mechanical ventilation were lower in the treatment group than in the control group.Mortality rate was significantly lower in the treatment group (0.696%) than the control group (4.724%).After treatment,NRS2002 score was positively correlated with mortality rate of COPD patients with malnutrition risks.Conclusion For the COPD patients with malnutrition risks,active nutritional intervention can improve their nutrition status (lower NRS2002 score),increase the number of respiratory muscles to alleviate anoxia,enhance cellular immune function,and thus improve their prognosis.

    ABSTRACT Objective To observe influence of danhong injection on levels of serum S100B protein and neuron-specific enolase (NSE) in patients with acute cerebral infarction (ACI),and to explore its clinical effect.Methods Eighty cases of ACI were randomly divided into treatment group and control group with 40 cases in each group.Both groups were given routine treatment according to the clinical guideline.Treatment group received danhong injection once a day for 14 days.Levels of serum S100B protein and NSE were detected 1,3,5 and 10 days after ACI.The patients in the two groups were evaluated by NIHSS 1 and 21 day(s) after ACI.Results Compared with control group,levels of serum S100B protein and NSE were significantly lower in treatment group than in control group 3 and 5 days after ACI (all P<0.01).NIHSS scores were significantly decreased 21 days after ACI in both groups,and NIHSS scores were lower in treatment group than in control group (P<0.05).Peak concentration of S100B protein in treatment group and control group was positively correlated with NIHSS scores (r=0.761 and r=0.792,respectively,both P<0.01).Conclusion Danhong injection can decrease serum S100B and NSE levels,and improve the neurological function in ACI patients.

    ABSTRACT Objective To explore the effect of sample-solvent compositions on the determination of Astragali Ⅳ by high performance liquid chromatograph- evaporative light scattering detector (HPLC-ELSD).Methods Radix astragali and ganduqing granules served as samples.Methanol,90%methanol,80%methanol,70%methanol,32%acetonitrile,15%acetonitrile,and water were sample solvents.HPLC-ELSD was used to determine content of astragaloside Ⅳ.Results The Results showed that the 90%methanol solution was an appropriate sample solvent with good system suitability,precision,accuracy,and,linearity etc..Conclusion This method benefits the quality control of astragaloside Ⅳ in Radix astragali and agents containing Radix astragali.

    ABSTRACT Objective To establish a method for the determination of hesperidin and cinnamaldehyde in siweiqing buccal tablets.Methods A high performance liquid chromatography (HPLC) method was established.The chromatographic conditions were as follows: the chromatographic column Inertsil ODS-SP(4.6 mm×150 mm,5 μm) with the mixture of acetonitrile and water as mobile phase in gradient mode,the detection wavelength of 290 nm,the flow rate of 1.0 mL&#8226;min-1,the column temperature of 30 ℃,the theoretical plate number no less than 3 000.Results The linear relationship between content and peak area was obtained in the range of 0.025-0.500 μg for cinnamaldehyde and 0.10-2.00 μg for hesperidin.RSD of the peak area of cinnamaldehyde and hesperidin for the precision test,the stability test and the repeatability test were 1.16%and 1.03%,1.27%and 1.08%,1.23%and 1.28%,respectively.The average recovery of cinnamaldehyde was 99.24%with RSD of 1.65%(n=9).The average recovery of hesperidin was 99.39%with RSD of 1.85%(n=9).Conclusion The method can be applied to quantitative assay of cinnamaldehyde and hesperidin in siweiqing buccal tablets with good reproducibility and specificity.The recovery result met the acceptance criteria.The method can effectively control the quality of siweiqing buccal tablets with good specificity,reproducibility and stability,and can be applied to the quality control of siweiqing buccal tablets.

    ABSTRACT Objective To determine the contents of geniposide in different parts of the fruit of Gardenia jasminoides Ellis with different maturity degree produced in fangxian,in order to find the optimum harvest periods and the medicinal parts.Methods The maturity of gardenia fruit are divided into four levels according to the color from totally green to yellow and red,and then the contents of geniposide were determined by HPLC.Results The contents of geniposide in all four levels of maturity reached the standard by the Chinese Pharmacopoeia.The content of geniposide in fruits of caesious color is up to 13.01%.Conclusion There is a negative correlation between the maturity and the content of geniposide in the mature fruit of Gardenia jasminoides Ellis.Caesious fruit has the best maturity degree.It is reasonable to use the whole fruit as medicine according to the Chinese Pharmacopoeia.
