    Objective To investigate potential protection and mechanism of glucocorticoid receptor(GR) agonist dexamethasone on renal ischemia reperfusion injury.Methods Eighteen male C57BL/6 mice were randomly divided into three groups: SHAM group(SHAM), kidney ischemia reperfusion injury group(IRI), and dexamethasone pretreatment group(DEX).The mice in IRI group and SHAM group received saline 1 h before ischemia(4 mg.kg-1, ip), But the mice in DEX group received dexamethasone(4 mg.kg-1, IP) 1 h before ischemia.The mice in the IRI and DEX groups were kept in 32 ℃ infant incubator for 1 h after clamping their renal pedicles, and then the right kidneys were removed.The mice in SHAM group underwent the same process, except for clamping the renal pedicles.These mice were sacrificed after 24 h of reperfusion.Blood samples were collected and the levels of serum creatinine(Cr) and urea nitrogen(BUN) were measured.The pathology of renal tissues was observed according to PAS staining, and the mRNA levels of MCP-1and IFN-γ were measured by PCR.Results Compared with the SHAM group, the levels of Cr and BUN in IRI group were significantly higher.Moreover, renal tubular necrosis, proteincast, inflammatory cell infiltration, and the mRNA level of MCP-1 and IFN-γ were all higher in IRI group than in the SHAM group.Conclusion GR agonist dexamethasone reduces renal ischemiareperfusion injury by inhibiting inflammation.

    Objective To study the characteristics of metabolism in rabbits with gallstone, and to explore the intervention by dahuanglingxian capsule for the disease.Methods Sixty New Zealand rabbits were randomly divided into 4 groups: normal control group, drug control group, model group and dahuanglingxian capsule group.The rats of the 4 groups were given normal diet, normal diet+dahuanglingxian capsule, lithogenic diet and lithogenic diet+ dahuanglingxian capsule, respectively.The course lasted 8 weeks.Plasma metabolism characteristics were analyzed by nuclear magnetic resonance hydrogen spectrum.Results Compared with the normal control group, gallstone was induced in 100% of rabbits in the model group(P<0.01).The plasma lactic acid, glutamine and malic acid were significantly increased, but glycine decreased with dysfunction of liver and kidney.The rate of gallstone formation in the dahuanglingxian capsule group was markedly reduced when compared with the model group(P<0.01).As compared to the model group, plasma glutamine and malic acid in the dahuanglingxian capsule group were lower, and glycine was higher with the improvement of liver and renal function.Conclusion Elevated plasma lactic acid, glutamine and malic acid and decreased glycine are the characteristics of metabolism in rabbits with gallstone.dahuanglingxian capsule reduces plasma glutamine and malic acid, and increases plasma glycine in rabbits with gallstone, implicating that dahuanglingxian capsule can effectively prevent and treat gallstone.

    Objective To study the components of Vernonia anthelmintica(L.) Wild in rat blood by serum fingerprinting analysis.Methods High performance liquid chromatograph method was used to establish the fingerprint of the herb.The ingredients into the blood were acquired by repeated silica gel column chromatography and Sephadex LH-20 gel column chromatography.Results Four compounds were isolated from the herb of Vernonia anthelmintica(L.) Wild.Prototypes of three compounds were absorbed into the blood: purple riveting hormone, 5,7,3’, 5’-tetrahydroxy flavanone, and 3’, 4’, 7-trihydroxy flavonoids.Conclusion Serum pharmacological method can be used to analyze the composition of herbs absorbed into the blood, and provide a reference to find the functionary composition of herbs in vivo.

    Objective To observe the protective effect of black yeast β1 ,3-1, 6 glucan in rats’ radiation injury.Methods Thirtyfive SD rats were randomly divided into six groups: control(C group); simple medicine administration(m group); 5 Gy radiotherapy(5 Gy group); 5 Gy radiotherapy with medicine(5 Gy+m group); 0.5 Gy radiotherapy(0.5 Gy group); 0.5 Gy radiotherapy with medicine(0.5 Gy+m group).Before irradiation, all the rats were given with the black yeast β1, 3-1, 6 glucan for two weeks except C group, 5 Gy group and 0.5 Gy group [1 g•(100 g)-1 by mouth, once daily].For groups receiving high dose radiation, the body weight, peripheral blood, serum SOD activity and MDA content before irradiation, and 3, 7 and 14 days after irradiation were detected.The bone marrow cell micronucleus and spleen index on the 14th day after radiotherapy was also measured.For groups of low dose radiation, DNA injury was evaluated with immunofluorescence focus number of peripheral blood lymphocytes 2, 4, 8 and 24 h after irradiation.Results The black yeast β1, 3-1, 6 glucan is effective in promoting weight recovery, reducing bone marrow suppression, delaying of SOD activity reduction and increasing MDA content after irradiation(the difference was statistically significant).Black yeast β1, 3-1, 6 glucan reduced the micronucleation rate, and the difference was statistically significant.It appeared to maintain the original size of the spleen to protect the immune system, and repair the DNA injury after low dose of irradiation.Conclusion The black yeast β1, 3-1, 6 glucan has some protective effect on SD rats with systemic radiation injury.

    Objective To study the antiinflammatory and antibacterial effect of paozhen bingdu tablet.Methods Mice were divided into 5 groups randomly: normal saline group, positive group, high, middle and low dose groups of paozhen bingdu tablet(10.2, 6.8, 3.4 g.kg-1, respectively).The antiinflammatory effect of paozhen bingdu tablet was observed via mouse earswelling model and cotton pellet induced granuloma and paw edema model, and by using mouse peritoneal capillary oozing test.The antibacterial effect of paozhen bingdu tablet was observed through in vitro and in vivo antibacterial test.Results The high dose of paozhen bingdu tablet significantly relieved earswelling, inhibited peritoneal capillary oozing, and reduced paw edema degree and cotton granuloma weight(P<0.05).Paozhen bingdu tablet had in vitro antibacterial effects on Staphylococcus aureus, Staphylococcus epidermidis, Shigella dysentery bacillus, Acinetobacter, Enterococcus, Pseudomonas aeruginosa, E.coli, Shigella flexneri, Klebsiella pneumoniae, Enterobacter cloacae, Candida albicans, B Streptococcus, and Pulmonary Streptococcus.The minimum inhibitory concentration(MIC) was 1.00, 1.00, 4.00, 4.00, 4.00, 4.00, 4.00, 4.00, 4.00, 4.00, 4.00, 4.00, 4.00 g.mL-1, respectively.Paozhen bingdu tablet reduced mortality in mice infected with Staphylococcus aureus and Staphylococcus epidermidis.Conclusion Paozhen bingdu tablet has anti inflammatory and antibacterial effects.

    Objective To determine and compare the protective effect of different concentrations of encephalon glycoside and ignotin injection on rat pheochromocytoma PC-12 cells and human neuroblastoma SH-SY5Y tumor cells.Methods The cell survival rates and the levels of NSE and BDNF in cellular media were detected by thiazole blue method(MTT) and ELISA respectively. Results Encephalon glycoside and ignotin injection within the polypeptide concentration range of 0.1-300.0 μg.mL-1 exerted protective effect on PC-12 cell injury induced by 0.000 34% H2O2, and the highest protective rate was(43.0±9.2)%.In the polypeptide concentration range of 0.1-30.0 μg.mL-1, encephalon glycoside and ignotin injection effectively protected SH-SY5Y cell injury induced by 0.000 34% H2O2, with the highest protective rate being(50.5±11.9)%.In ELISA, NSE in both SH-SY5Y and PC-12 cell culture media was significantly decreased by encephalon glycoside and ignotin injection, while BDNF was significantly increased.Conclusion Encephalon glycoside and ignotin injection has a protective effect on H2O2 induced PC-12 and SH-SY5Y cell injury.NSE is down regulated during the treatment process.The upregulation of BDNF may be involved in the mechanism of protection of encephalon glycoside and ignotin injection on the cells.

    Objective To evaluate the efficacy and safety of alefacept in the treatment of patients with psoriasis.Methods The randomized controlled trials(RCT) on alefacept in the treatment of the patients with psoriasis were collected by searching The Cochrane Library(issue 3, 2012), specialized trials registered in Cochrane Skin Group(2012), MEDLINE(1966-2012), and Embase(1974-2012).The quality of the trials was assessed according to the Jadad scale by two independent reviewers.RevMan 5.0 software was used for statistical analysis.Results Six RCTs involving 1 289 patients were included in the analysis.All patients were at least 16 years old diagnosed with chronic moderate to severe plaque psoriasis.Significant improvement was achieved in patients receiving alefacept 0.075 mg•kg-1•w-1 or 7.5 mg•w-1 intravenously, or 10 mg•w-1 or 15 mg•w-1 intramuscularly for 12 weeks as compared with placebo groups.After treatment for 12 weeks, the mean scores of psoriasis areaandseverity index(PASI) continued to decrease with improvements in quality of life(QOL).Conclusion Alefacept therapy for 12 weeks result in significant improvements in adult patients with moderate to severe psoriasis.More RCTs are required to determine the optimal dose for psoriasis.Alefacept Is well tolerated and provided long lasting remission of psoriasis.

    Objective To observe the effects of jingfang zhiyang granules on experimental itchy skin models.Methods The incubation period and the times of itching in mice pruritus model induced by dextran, and guinea pig pruritis model induced by histamine phosphate were researched.The inflammatory edema of ear induced by xylol in mice and the wet weight, dry weight of granuloma induced by sterilized cotton ball were also investigated. Jingfang zhiyang granules were administered intragastrically for 7 days at concentrations of 20.28, 10.14, 5.07 g.kg-1(raw medicinal herbs) in mice,12.08,6.04, and 3.02 g.kg-1 in guinea pigs, and 14.04,7.02, and 3.51 g.kg-1 in rats.Results All dosages of jingfang zhiyang granules could decrease the times of itching.The middle dose of jingfang zhiyang granules prolonged the incubation period in mices.The high and middle doses of jingfang zhiyang granules inhibited local itching in guinea pigs and decreased ear swelling in mice.All dosages of jingfang zhiyang granules decreased the wet weight and dry weight of granuloma in rats.Conclusion Jingfang zhiyang granules demonstrated antipruritic and anti inflammatory effects on experimental pruritus animal models.

    Objective To investigate the safety of sanhuang sanyu cataplasm. Methods Six rabbits were randomly divided into 2 groups.Skin irritation of sanhuang sanyu cataplasm on rabbits’ complete and injured skin was observed.Thirtysix rabbits were randomly divided into 6 groups, acute toxicity of sanhuang sanyu cataplasm on injured and injured skin of rabbits was examined.Thirty guinea pigs were randomly divided into 3 groups.Allergic reaction from sanhuang sanyu cataplasm in guinea pig skin was observed.Results Sanhuang sanyu had no irritation and acute toxicity on complete or injured skin of rabbits.There was no allergic reaction in guinea pig skin.Conclusion Sanhuang sanyu cataplasm is safe and nontoxic.

    Objective To explore the influence of gastrodin sustained release tablets on CD+4CD+25 regulatory T cells(Treg) and interleukin-17(IL-17) in patients with highrisk transient ischemic attack(TIA) in posterior circulation.Methods Sixtythree patients with highrisk transient ischemic attack in posterior circulation were randomly divided into control group(n=31) and experiment group(n=32).The control group was conventionally treated with aspirin entericcoated tablets, atorvastatin calcium tablets and salvianolate injection and so on.The experiment group was treated with gastrodin sustained release tablets plus conventional therapy. Both groups were treated for 28 days before evaluation.The frequencies of Treg and the concentration of IL-17 in the peripheral blood were detected by flow cytometer and enzymelinked immunoadsordent assay before and after the treatment.Results The frequencies of Treg and the concentration of IL-17 had no significant differences between the two groups before the treatment(P>0.05).The frequencies of Treg were found to be significantly higher in experiment group(15.30±2.97) than in control group(13.48±2.58)(P<0.05), while the concentration of IL-17 was found to be significantly lower in experiment group(184.63±65.76) than in control group(231.91±68.30)(P<0.05) after the treatment.Conclusion The gastrodin sustained release tablets may help to correct the imbalance in autoimmune inflammatory response in patients with highrisk TIA in posterior circulation.

    Objective To compare the clinical effect of orally administered N-acetylcysteine(NAC) combined with intravenous sodium bicarbonate or combined with intravenous saline in preventing contrastinduced nephropathy(CIN) in patients without diabetes mellitus after coronary or peripheral angiography.Methods One hundred and twenty patients were randomly assigned to receive oral NAC combined with intravenous 1.5% sodium bicarbonate(SB group, 62 patients) or oral NAC combined with intravenous normal saline(NS group, 58 patients) before angiography.NAC was given at the dose of 1 200 mg bid on the day of and the day before angiography.There were no significant differences in age, sex, BMI, hypertension, Scr, serum electrolytes, arterial blood gas or contrast volume between the two groups.Scr was assessed right before and 48 and 72 hours after contrast administration.All patients received administration of hypotonic nonionic contrast.The primary endpoint was CIN.Results CIN developed in 2 patients(3.23%) in the SB group and in 8 patients(13.79%) in the NS group.The incidence of CIN was significantly lower in the SB group as compared with the NS group(P<0.05).Conclusion Oral NAC combined with intravenous sodium bicarbonate may be superior to oral NAC combined with intravenous saline in preventing CIN in patients without diabetes mellitus undergoing coronary or peripheral angiography.

    Objective To evaluate safety and efficacy of dexmedetomidine in postoperative patientcontrolled intravenous analgesia(PCIA) for gynecologic laparotomy patients.Methods Eighty patients(18-65 years of age, ASA ClassⅠor Ⅱ) undergoing abdominal hysterectomy or myomectomy, were randomly divided into two groups: dexmedetomidine combined with fentanyl group(Group DF, 40 cases) and fentanyl group(Group F, 40 cases).The patients in Group DF received fentanyl 10 μg•mL-1 plus dexmedetomidine 2 μg•mL-1, and the patients in Group F received fentanyl 10 μg•mL-1 for PCIA.Cumulative PCIA requirements, Visual analogue pain score(VAS), Remesay sedation score(RSS), Vital signs(heart rate, blood pressure, respiratory rate, SpO2), post operative quality of recovery score(QoR9), and PCIA related adverse events were recorded for 24 h after operation.The Mini Mental State Examination(MMSE) score was evaluated to assess the cognitive function before and after the operation.Results The heart rate of patients in Group DF stablized within 1 h after operation, and the mean arterial pressure returned to the preoperative level 4 h after operation; yet the heart rate and mean arterial pressure in Group F remained elevated 24 h after operation(P<0.05).Compared with Group F, the doses of fentanyl in Group DF were significantly decreased(P<0.05).There was no significant decrease in MMSE scroe 1 day after operation.Conclusion Dexmedetomidine combined with fentanyl in PCIA may be the effective and safe for gynecologic laparotomy patients.

    Objective To evaluate the protective effects of nicorandil on myocardium and microvasculature after intervention therapy in patients with acute ST-segment elevation myocardial infarction.Methods All 120 patients with acute ST-segment elevation myocardial infarction who received percutaneous coronary intervention were randomly divided into two groups.Patients in the control group received standard therapy, and patients in the treatment group received standard therapy plus oral nicorandil 5 mg daily for 6 months.During the follow up, wall motion score index(WMSI), left ventricular ejection fraction(LVEF), left ventricular end diastolic volume(LVEDV) were determined and recorded by echocardiography.The myocardial infarction area was measured via SPECT.Episodes of recurrent angina pectoris were recorded.Results The recovery rate for the abnormal movement of the ventricle segments in the treatment group were significantly higher than that of the control group(68.8% vs.51.2%, P<0.05) after 6 months.LVEDV was decreased and LVEF increased significantly in the treatment group as compared with that of the control group, respectively(P<0.05).The WMSI and the myocardium acquire isotope score index of the treatment group decreased significantly as compared with that of the control group(P<0.05).The episodes of recurrent angina pectoris in treatment group were less than those of control group significantly(P<0.05).Conclusion Nicorandil can decrease infarction area after PCI in acute STEMI, and prevent left ventricular remodeling.

    Objective To evaluate the efficacy and safety of gatifloxacin in the treatment of ordinary type typhoid fever.Methods In the comparative study, patients were enrolled and randomized to two groups(test group and control group).The patients received oral gatifloxacin(test group) or intravenous levofloxacin(control group) 400 mg once daily for 14 days.Cure rate and clinical efficacy rate [median(95% confidence interval) fever clearance time and relapse rate] were compared.The incidence of adverse reaction was evaluated.Results It showed that the cure rate and clinical efficacy rate in the test group were 81.8% and 90.9% in the test group, and 83.3% and 91.7% in the control group, respectively(P>0.05).The median(95% confidence interval) fever clearance time of gatifloxacin and levofloxacin were 182.0(155.5-208.5) hours and 162.1(136.6-187.6) hours, respectively(P>0.05).The relapse rate in the test and control group was 9.1% and 12.5%, respectively(P>0.05).The incidence of adverse reaction in the test and control group was 9.1% and 8.3%, respectively(P>0.05).Conclusion Gatifloxacin has an excellent clinical efficacy and good safety profile in treating patients with ordinary type typhoid fever.

    Objective To establish a method for determining schizandrin, schisantherin A, deoxyschizandrin and γ-schishandrin in Schisandra chinensis by RPHPLC, and to study lignans composition of Schisandra chinensis from different origins.Methods A Venusil XBP C18 column(4.6 mm×250 mm, 5 μm) column was used with gradient elution by methanol(A)water(B)(0-25 min, 72%A; 26-50 min, 77%A) as the mobile phase.The flow rate was 1.0 mL•min-1.The column temperature was 25 ℃.The detection wavelength was set at 250 nm.Results The calibration curves of schizandrin, schisantherin A, deoxyschizandrin and γ-schishandrin were linear within the ranges of 1.27-10.17 μg(r=0.999 6), 0.27-2.13 μg(r=0.999 0), 0.32-2.52 μg(r=0.999 5), and 0.89-7.19 μg(r=0.999 4), respectively,.The average recoveries were 100.51%(RSD of 2.09%), 101.20%(RSD of 1.83%), 102.05%(RSD of 1.99%), 99.73%(RSD of 1.34%), respectively.Conclusion The method is accurate, simple and selective, providing a basis for improvement of quality evaluation for Schisandra chinensis.

    Objective To establish a headspace gas chromatography method for determination of alcohol residue in qingkailing injection.Methods The alcohol residue in qingkailing injection was separated by HPINNOWAX capillary column with an FID detector.The carrier gas was nitrogen.The flow rate was 4.0 mL.min-1.The injector temperature and the detector temperature were controlled at 200 ℃ and 250 ℃, respectively.The splitting ratio was 2: 1.The programmed column temperature was set as follows: maintained at 60 ℃ for 8 min and then increased to 180 ℃ followed by holding for 8 min.The sample was dissolved in water and dioxane was used as the internal standard.The containers of headspace injector were in equilibrium at 90 ℃ for 20 min.Results The detected solvents were separated completely.The calibration curve of solvent showed a good linear relationship(r=0.999 98) with good accuracy.The average recovery rate was 99.51% with the RSD of 0.74%.Conclusion The established method was convenient and repeatable with simple operation and accurate Results It can be used for determination of alcohol residue in Qingkailing injection.

    Objective To study folateconjugated chitosan oligosaccharide nanoparticles(FCS-PLGA-NPs) targeting to tumor cells via folate receptormediated endocytosis.Methods The carboxyl group of folic acid was activated by EDC.The activated folic acid was then conjugated to the amino groups of chitosan oligosaccharide to obtain folateconjugated chitosan oligosaccharide(F-CS).Carboxylterminated PLGA was used to prepare paclitaxelloaded nanoparticles(PLGA-NPs).F-CS-PLGA-NPs was prepared by the reaction of the amino groups of F-CS and the carboxyl groups of PLGA activated by EDC.Ultraviolet and infrared spectrometry was used to identify F-CS and F-CS-PLGA-NPs.Folate coupling rate was determined by ultraviolet spectrometry.The morphology of F-CS-PLGA-NPs was observed by SEM.Particle size and Zeta potential were measured by laser particle size analyzer.Drug loading was determined by HPLC.Results The folateconjugated chitosan oligosaccharide was synthesized successfully and the PLGA nanoparticles were successfully modified with folateconjugated chitosan oligosaccharide.Folate coupling rate of F-CS and F-CS-PLGA-NPs was 24.39% and 8.87%, respectively.Particle size of F-CS-PLGA-NPs, was(321.00±0.76) nm and Zeta potential was (22.60±0.26) mV.Drug loading was(5.10±0.25)%.Conclusion The preparation process of F-CS-PLGA-NPs is feasible.

    Objective To establish a RP-HPLC gradient elution method for determination of uridine, guanosine and adenosine in Pinellia ternate.Methods The extracts were separated on a ZORBAX-SB-C18 analytical column(4.60 mm×150 mm,5 μm) with a solvent system containing CH3OH-H2O, the flow rate at 0.6 mL.min-1 and the column temperature at 25 ℃.The analytes were detected at 260 nm UV wavelength.Results The linear ranges of uridine, guanosine and adenosine were 0.078 4-0.784 0 μg(r=0.999 5), 0.079 8-0.798 0 μg(r= 0.999 5), and 0.085 2-0.852 0 μg(r=0.999 4),respectively.Conclusion This method has a good reproducibility, which can be used for quality control of Pinellia ternate.

    Objective To establish an HPLC-UV method for the determination of the berberine hydrochloride and palmatine hydrochloride in SSYHG and studies on its fingerprint.Methods HPLC gradient elution was applied for the fingerprints of SSYHG.The contents of berberine hydrochloride and palmatine hydrochloride in SSYHG were measured.The column was GenimiC18 column.Mobile phase was acetonitrile(A) -0.01 mol.L-1 ammonium acetate(B).Detection wavelength was 345 nm.Results The fingerprint of SSYHG was established;the relative retention time and the relative area of the common peaks were determined;the similarity of ten batches of SSYHG samples was above 0.953 7.The average recoveries of berberine and palmatine hydrochloride were 97.52% and 96.72%, respectively.Conclusion The method is accurate, reliable and reproducible.It can be used to control the quality of SSYHG.

    Objective To establish and compare Raman spectral fingerprints of different yam cultivars and provide a methodological basis for the quality control of yam.Methods Twenty different cultivar batches of yam were collected from Henan, Guangxi, Guangdong and Wenzhou.Their fingerprints were established by confocal microRaman spectroscopy at 785 nm.The component of main peaks was identified by the characteristic Raman frequency and the initial specified table of compounds.Matlab fingerprint analysis software, statistical Methods , principal component analysis and derivation method were used to analyze the similarity of different batches.Results The Raman fingerprints of different yam cultivars were significantly different.The similarity of yam from Henan, Guangxi and Guangdong was 0.95, while the similarity of batches from Wenzhou was as low as 0.82.Dioscorea alata was distinct from the other three different cultivars(Dioscorea opposite, Dioscorea persimilis and Dioscorea fordii) in terms of the reference index number, displacement, height and area of the characteristic peak.Conclusion Raman spectroscopy can distinguish Dioscorea alata from the other three different yam cultivars, and be used as a quality control method for Dioscorea alata.
