    ABSTRACTObjectiveTo study the rat intestine absorption characteristics of protopine (Pro)and tetrahydropalmatine in extracts of Rhizoma Corydalis decumbentis.MethodsThe isolated everted intestinal sac was utilized to study the intestine absorption characteristics of protopine and tetrahydropalmatine.The sample’s concentration was determined by HPLC method.The apparent permeability coefficients (Papp)and the mean cumulative permeation amount versus time curves were calculated and drawn.The Papp of protopine and tetrahydropalmatine was investigated and compared under different everted gut sacs, different concentrations and different pH values.ResultsThe Papp of protopine and tetrahydropalmatine in ileum was about 2.58×105 cm•s1 and 3.15×105 cm•s1, respectively, and the difference was remarkable compared to other intestine sacs (P<0.05).The Papp was decreased as Pro level increased,,and the cumulative permeation amount was nonlinear increased in the duodenum, jejunum, and ileum.When the pH value decreased from 7.4 to 5.4, the absorption of protopine and tetrahydropalmatine across intestinal membrane presented declining tendency.ConclusionThe absorption of protopine and tetrahydropalmatine in ileum is much better than that in other parts of intestine.The pH values could affect intestinal absorption of protopine and tetrahydropalmatine.The absorption of protopine in duodenum, jejunum and ileum might be saturated.

    ABSTRACTObjectiveTo study the effect of deguelin on genes expression in COX/EP pathway of zebrafish embryos.MethodsRealtime quantitative PCR detecting system was used to compare genes expression pre and posttreatment of deguelin.ResultsCompared with the control, the expression on transcript levels of ptges, ptges3a, ep4a, ep4b, ep2a, ep2b, egr1, cyclin D1 in COX/EP pathway were decreased to 0.46±0.16,0.82±0.06,0.59±0.03,0.12±0.03,0.15±0.04,0.16±0.01,0.19±0.08,0.33±0.06,respectively(P<0.01 or P<0.05).ConclusionDeguelin arrests zebafish embryonic development at sphere stage, which may be related to the inhibition of main genes expression in the COX/EP signaling pathway.

    ABSTRACTObjectiveTo investigate the effects and mechanism of lianqixiaoke capsules (LXC) on advanced glycation endproducts(AGEs) and the oxidative stress injury in kidneys of STZinduced diabetic rats.MethodsThe Wistar rats were randomly divided into 6 groups, which were the control group fed with chow food, the other groups given with high fat diets.After 4 weeks, the rats except the control were intraperitoneally injected with streptozotocin (STZ)30 mg•kg1.The diabetic rats were treated with LXC at high, middle and low doses, metformin and vehicle, respectively.Eight weeks later, the blood glucose,UALB,SOD,MDA, AGEs,TGFβ1 and TNFα were measured.And the kidney was removed for pathological analysis.Results In the LXC group, FBG was(18.67±2.52) mmol•L1, MDA was(1.78±0.39) nmol•g1, UmALB was (41.84±8.23) μg•L1,AGEs was (116.91±19.96) ng•mL1, TGFβ1 was (33.22±9.94) ng•mL1,TNFα was (54.01±17.55) pg•mL1,all of which were declined and SOD was(190.11±15.88) U•mg1,which was elevated.The function and pathological changes in kidney were significantly improved compared to those in the control group, especially for the high dose of LXC (P<0.05).ConclusionThe model of diabetic nephropathy in diabetic rats is successfully set up by feeding fat diet and injecting low dose of STZ in rats.LXC decreases AGEs and relieves diabetic kidney injury, the mechanism of which could be associated with inhibiting production of AGEs, lowering oxidative stress and inflammatory factors.

    ABSTRACTObjectiveTo investigate effects of taorenhonghua decoction on rats with vascular dementia induced by chronic global ischemia.MethodsPermanent occlusion of the bilateral carotid arteries was established in rats.The treatment group was daily orally administered with taorenhonghua decoction after surgery.Rats in sham and model groups were treated with equalvolume of saline daily.Morris water maze test was conducted on day 30 postsurgery.ResultsRats in model group [(33.4±3.9) s] consistently took longer to find the hidden platform than those in sham group (20.1±2.5) s (P<0.05).The prolonged escape latency in treatment group (24.5±3.1)s was significantly shortened by longterm administration of taorenhonghua decoction (P<0.05).In the spatial probe trials, the rats in model group spent more time(37.5±3.3) s on searching the target quadrant where the platform was located than the sham group(20.3±4.0) s (P<0.05).The rats in treatment group spent less time (29.1±3.6) s in searching the platform in the target quadrant than the model group did, and the significant difference was observed between model and treatment groups (P<0.05).Compared with shamoperated group, MDA levels were significantly increased, while SOD and catalase activities were significantly decreased in model group, while which were reversed by longterm administration of taorenhonghua decoction in treatment group.HE staining showed neuronal loss, shrinkage and marked vacuolar changes in the hippocampus of model group, which was significantly attenuated by taorenhonghua decoction.ConclusionTaorenhonghua decoction can improve the study and memory ability of rats with vascular dementia induced by chronic global ischemia, which may partly be related to its antioxidant actions.

    ABSTRACTObjectiveTo evaluate in vitro release behavior of fexofenadine capsules, establish a LCMS/MS method for the content determination of fexofenadine and study its pharmacokinetics in Beagle dogs.MethodsThe release behavior was evaluated by using a cup method in four media;a LCMS/MS method was used to determine drug concentration after a single dose of administration of tablets or capsules in six dogs;pharmacokinetic parameters were calculated with the DAS2.1.1 program.ResultsThe two preparations showed good release behavior;the AUC0t of fexofenadine capsules and tablets were (20.00±7.48)and (16.42±9.37) mg•L1•h, AUC0∞ were (20.30±8.08)and (16.68±9.44) mg•L1•h, t1/2 were (3.10±1.60)and (4.54±2.03)h, tmax were (1.58±0.49)and (1.67±0.68) h, and Cmax were (4.24±1.69)and (3.68±1.89) mg•L1, respectively.ConclusionThe capsules and tablets of fexofenadine present similar release behavior in vitro.No obvious difference has been found between the main pharmacokinetic parameters of them, which suggests they would have the same therapeutic efficacy.

    ABSTRACTObjectiveTo study the chemical constituents of volatile oils from barks of Zanthoxylum planispinum Sieb.et Zucc.MethodsThe volatile oil were extracted by steamstilling and identified by GCMS.The percentage composition of volatile oil was calculated according to the peak area normalization method.Results13 compounds were identified, in which the major constituents were 2undecanone (34.78%), 2tridecanone (10.94%), vinyl decanoate(9.43%)and αbisabolol(7.41%).ConclusionThe volatile oils from barks of Zanthoxylum planispinum Sieb.et Zucc.growing in Zhejiang province is rich in 2undecanone and worth being developed.

    ABSTRACTObjectiveTo investigate the effect of genistein on cell proliferation, apoptosis and cell cycle of human cervical cancer cell line Siha. MethodsSiha cells were incubated with 50 mol•mL1 genistein diluted in DMSO or DMSO at equal volume.The cell growth was measured by MTT.Cell apoptosis, cell cycle and expression of cyclin A and cyclin B were detected by means of flow cytometry (FCM). ResultsThe proliferation of Siha cells treated with genistein was significantly inhibited compared to the control group (P<0.05);FCM assay showed that cell apoptosis was increased by being treated with genistein for 48 h (67.45±1.50)% compared to that in the control (45.37±1.65)% (P<0.01);cells arrested at G2/M phase were increased from (9.55±0.71)% to (79.54±1.09)% (P<0.01);the protein expression of cyclin A and cyclin B were upregulated from (1.84±0.11)% to (37.38±0.71)% (P<0.01)and from (69.44±1.84)% to (97.70±1.20)% (P<0.01), respectively. ConclusionGenistein inhibits the growth of cervical cancer cell line Siha in vitro, and the mechanism of which is likely due to upregulation of cyclin A and cyclin B and arresting cell cycle at G2/M phase.

    ABSTRACTObjectiveTo evaluate the efficacy and safety of sertaconazole nitrate suppositories for treating vulvovaginal candidiasis.MethodsA doubleblind, randomized controlled trial was conducted.30 cases in both treatment group and control one were applied with sertaconazole nitrate suppositories and clotrimazole ones for 1-2 times, respectively.Patients were evaluated at day 7±1 and day 28-35 after ceasing medication. Results28 cases in the medical trial group and 30 cases in control were evaluated;The clinical efficacy at day 7±1 were 89.29% in the test group and 86.67% in control group;and 81.48% versus 66.67% at day 2835 after drug withdrawn, respectively.No treatmentrelated adverse events occurred in both groups.ConclusionThis study suggests that sertaconazole nitrate suppositories are effective and safe for the treatment of vulvovaginal candidiasis.

    ABSTRACTObjectiveTo study the efficacy and safety of dinoprostone suppository for odinopoeia.Methods62 cases of fullterm singleton pregnant women with fetal head down had odinopoeia indications without using dinoprostone suppository were conventionally placed in the vagina with 10 mg dinoprostone suppository.The cervix uteri was observed after 12 and 24 h, uterine contraction and duration to labor, the fetal heart rate changes, amniotic fluid delivery, cesarean signs and adverse drug reactions were detected.ResultsThe dinoprostone suppository obviously promoted cervical ripening with Bishop score 12 h after medication as (7.23±2.25), and 24 h as (9.76±3.02).The dinoprostone suppository made high abortion rate and was safety for mother and child with few adverse reaction.ConclusionDinoprostone suppository is simple to use, effective, high security, controllable.It increases opportunities for natural labor, reduces caesarean section rates, which is a safe cervical ripening enhancer.

    ABSTRACTObjectiveTo evaluate the efficacy and toxicity of paclitaxel liposome plus nedaplatin in the treatment of advanced nonsmall cell lung cancer(NSCLC).Methods17 cases of NSCLC treated with paclitaxel liposome and nedaplatin for 2 to 6 cycles, by infusing paclitaxel liposome 135 mg•(m2)1 for 3 h on day 1 and combining nedaplatin 80 mg•(m2)1 as infusion on day 2.ResultsAmong 17 patients being evaluated for response to treatment, 1 showed complete response(CR), 4 showed partial response (PR), 3 showed stable disease (SD), 9 showed progress disease(PD).The main side reaction was haematological toxicity, including neutropenia and thromboeytopenia.The nonhaemutological toxicity was nausea and vomiting, mild liver injury, and fair loss.ConclusionPaclitaxel liposome plus nedaplatin was effective and well tolerated for treating patients with advanced nonsmall cell lung cancer.

    ABSTRACTObjectiveTo investigate the efficacy of sulfotanshinone sodium injection on patients with primary nephrotic syndrome(PNS).Methods83 patients with PNS were randomly divided into therapeutic group and control group, the latter was given routine therapy as prednisone 1 mg•kg1•d1 and other symptomatic treatment, the former was treated with sulfotanshinone sodium injection (50 mg and 5% glucose solution 250 mL intravenous drip once a day)based on the routine therapy for 3 weeks.Ddimer,prothrombin time (PT), activated partial thromboplastin time(APTT), 24hour urine protein, serum albumin, triglyceride(TG)and cholesterol(TC) were detected.ResultsAfter treatment, urine protein decreased to(2.48±0.79) g, blood plasma albumin increased to(37.56±3.21) g•L1, blood lipid declined to(TC 8.02±2.11 mmol•L1,TG 1.78±0.72) mmol•L1,PT elevated to(13.28±2.50) s, APTT increased to(33.42±5.98) s,Ddimer dropped to(0.93±0.49) mg•L1, whole blood viscosity lowered to(4.13±0.47) mPa•s, and blood plasma viscosity decreased to(1.39±0.54) mPa•s in the therapeutic group.The blood biochemical indicators in the treatment group were significantly improved compared to those in the control group (P<0.01).ConclusionSulfotanshinone sodium injection obviously ameliorates hypercoagulable state of PNS,decreases proteinuria and blood lipids, increases plasma albumin,and delays the progress of PNS.

    ABSTRACTObjectiveTo assess the clinical efficacy of celecoxib combined with eperisone hydrochloride for treating acute lumbar muscle sprain.MethodsAmong 72 cases with acute lumbar muscle sprain, 32 were treated with celecoxib 400 mg bid at the first day, and then with celecoxib 200 mg bid for 6 days;40 were treated with celecoxib combined with eperisone hydrochloride 50 mg tid for 7 days.The clinical effect and the adverse drug reaction at the 7th and 14th day were recorded.ResultsAt the 7th day, the excellent and good therapeutic rate was 90.0% in the experimental group and 75.0% in the control group(P<0.05).While at the 14th day, the excellent and good therapeutic rate was 95.0% in the experimental group and 93.8% in the control group(P>0.05).ConclusionThe clinical effect of celecoxib combined with eperisone hydrochloride would be more effective than monotherapy for acute lumbar muscle sprain in early period;there are no significant differences between the two treatments in later period.

    ABSTRACTObjectiveTo establish an extracting process for high purity αterpineol from pine nodular branch and study its stability.MethodsVolatile oil was extracted by steam distillation, and purified with silica gel column by eluting with petroleum ether:ethyl acetate (20:1)eluent.High temperature and strong light shining was used to test the stability of αterpineol.ResultsThe purified αterpineol was over 95.0%, which should be stored away from high temperature and strong light.ConclusionThe method is reliable, simple and suitable for αterpineol extraction from pine nodular branch.

    ABSTRACTObjectiveTo establish the quality standards for Artemisia rupestris L.from Xinjiang province by determining water content, total ash, crude material identification and rupestonic acid quantification.MethodsMoisture and total ash were determined based on the procedures recorded in the Appendix Ⅸ H, Ⅸ K of pharmacopoeia, published in 2005.And the Artemisia rupestris L. was identified by using a thin layer chromatography (TLC)method (the Appendix Ⅵ B).and the bioactive component rupestonic acid was determined by HPLC (the Appendix Ⅵ D), respectively.ResultsThe water contained in Artemisia rupestris L.was less than 8.0%, and the total ash was lower than 16.0%.The TLC spots were clear and specific;the rupestonic acid showed a good linear relationship at a range of 5.60-56.00 μg, with the regression equation as Y= 8.522 3X-6.1507, r= 0.997 2 respectively.And the average recovery was 99.7% (RSD=0.25%).ConclusionThe method is easy, accurate, quick and reproducible, which is benefit for the quality control of Artemisia rupestris L., and providing scientific basis for development of Artemisia rupestris L.

    ABSTRACTObjectiveTo determine the content of emodin in yijingling oral solution. MethodsHPLC was used with the mobile phase consisted of methanol0.1% methanoic acid (80:20);with the flow rate at 1.0 mL•min1;column temperature at 30 ℃ and detection wavelength at 254 nm.ResultsLinear range of emodin determination was 0.05-64.00 μg•mL1(r=0.999 9).The mean recovery was 99.55%, and RSD was 1.68%(n=9).ConclusionThe method is convenient, specific and reproducible for emodin determination in yijingling oral solution.

    ABSTRACTObjectiveTo investigate different wine processing technology on content of salvianolic acid B in danshen and screen out the best processing technology.MethodsThe four species of biennial Tomb Salvia as the original herb, wine stewing, wine fried, wine stewed one were brewed, extracted, and salvianolic acid B in which was detected by high performance liquid chromatography.ResultsThe comparison of Salvianloic acid B in the original herb, wine stewing, wine fried, and wine stewed one was 6.43%>4.66%>2.73%>1.85%, respectively.The loss of Salvianloic acid B in wine broiled, wine steamed, and wine stewed products was 27.5%,57.5%,71.2%, respectively. ConclusionWine processing enhances the efficacy of Salviamiltiorrhiza on eliminating cold and leading other medicines to targets and descends salvianolic acid B. Wine broiling is the optimum process.

    ABSTRACTObjectiveTo establish a method to determine the contents of ginsengnoside Rg1, ginsengnoside Re and ginsengnoside Rb1 in Radix Panax quinque folium L. by rapid resolution liquid chromatography(RRLC).MethodsZOBAX SBC18 column(4.6 mm×50 mm,1.8 μm)was used with the mobile phase consisting of acetonitrile(A) 0.1% phosphoric acid solution (B)with gradient elution(0-25 min,5%A→20%A;-35 min,20%A→40%A)at a flow rate of 2.0 mL•min1;the wavelength of detector was 203 nm and column temperature was set at 35 ℃.ResultsThe calibration curves showed good linearity in the range of 0.12-2.49 μg (ginsenoside Rg1),0.61-12.15 μg (ginsenoside Re)and 1.42428.48 μg (ginsenoside Rb1), with correlation coefficient as 0.999 8,0.999 6,0.999 9,respectively.The average recovery (n=6)was 97.8%,98.1% and 98.3%, and RSD was 1.0%,0.8% and 0.4%, respectively.ConclusionThis method is quick, accurate and reproducible, which is fit for quantification of Radix Panax quinque folium L.

    ABSTRACTObjectiveTo establish a simultaneous determination for alantolactone and isoalantolactone in guoxu a decoction by gas chromatography.MethodsAlantolactone and isoalantolactone of Radix Inulae Heleni in decoction of guoxu a were determined by gas chromatography, based on the optimized extraction methods.Results The linear range of alantoactone and isoalantolactone was 0.1-1.0 mg•mL1 (n=5).The mean recovery for alantolactone was 97.25%(n=6), RSD=0.93%.The mean recovery for isoalantolactone was 97.42%(n=6), RSD=0.90%.ConclusionThe result indicates that the method is sensitive and accurate,which can be used to simultaneousy determine alantolactone and isoalantolactone of Radix Inulae Heleni in decoction of guoxu a.
