    The circumstances fulfilling medicine supply mission for the Wenchuan earthquake medical rescue were retrospected. The achievements and deficiency of medicine supply in medical rescue were analyzed and some suggestions on emergent medicine supply in momentous emergency events were proposed.

    Objective To investigate whether the pregnantlevel estrogen induce the expansion of CD+4CD+25 regulatory T cells and its suppression activity in vitro. Methods① CD+4CD+25regulatory T cells labelled with CFSE were stimulated with antiCD3 and antiCD28 mAbs or PBS in the presence and absence of pregnant level estrogen (E2) (experimental group, control group A and control group B), or no any stimulus instead of PBS (placebo group). ②CFSElabelled CD+4CD+25 naive T cells were cultured with CD+4CD+25 regulatory T cells, which were pretreated for 2 hours with estrogen or not, and stimulated with antiCD3 and antiCD28 mAbs or PBS (experimental group, control group A and control group B). As placebo group, CFSElabelled CD+4CD+25 naive T cells were treated only with PBS. After 120 hours, the proliferation of labelled cells was detected by FACS and analyzed by Modfit LT soft. ResultsStimulated by antiCD3 and antiCD28 mAbs in 120 h, the proliferation index (PI) of CD+4CD+25regulatory T cells in experimental group were significantly higher than that in two control groups and placebo group. The proliferation of CFSElabelled CD+4CD+25 naive T cells mediated with antiCD3 and antiCD28 mAbs was inhibited by CD+4CD+25regulatory T cells. estrogen pretreated CD+4CD+25regulatory T cells could further enhance this effect. Conclusion It suggested that high level of estrogen in gestation can induce the expansion of CD+4CD+25 regulatory T cells and enhance its suppression, and may play a pivotal role in the maternalfeto tolerance.

    Objective To investigate the antivirus activity and mechanism of 13 extracts from 7 medicinal plants of the Selaginella genus on herpes simplex virus-Ⅰ (HSV-Ⅰ) in vitro. MethodsVero cells infected by HSV-Ⅰ were cultured with different extracts at different concentration for 72 hours , and the inhibitory effect on HSV-Ⅰ replication were evaluated by cell survival rates according MTT assay. Results In the Vero cell system, some of the extracts inhibited cytopathic effect(CPE) of cells infected with HSV-Ⅰ, and increased survival rates of cells. ConclusionSeveral Selagenella plants could be taken into account as new resource plants with antiHSV-Ⅰ action.

    Objective To observe the inhibitory effect of Abrus cantoniensis Hance on HBsAg and HBeAg in vitro. Methods The ethanol extracts of Abrus cantoniensis Hance was mixed with HBsAg and HBeAg masculine serum which had same capacity and same titer(the titer of 1:64).After mixing, the densities of Abrus cantoniensis Hance were 96,48,24,12 and 6 g•L-1,respectively. And PBS with masculine serum (the titer of 1:64) was made as blank control. The optical density (A)of HBsAg and HBeAg in each serum sample were determined by ELISA after 8 h and 16 h’s cultivation in water at 37 ℃. Results The ethanol extracts of Abrus cantoniensis Hance had obvious inhibitory effect on HBsAg and the HBeAg in serum at the density of 96 g•L-1 and 48 g•L-1 (P<0.05 or P<0.01), respectively. Conclusion Abrus cantoniensis Hance could inhibit obviously HBsAg and HBeAg in vitro.

    Objective To develop a simultaneous determination of dissociation constant values (pKa) of sulfamethoxazole (SMZ), sulfadiazine (SD) and trimethoprim (TMP). Methods Capillary electrophoresis (CE) conditions: a fused silica capillary column (60.2 cm×75 μm, effective length 50 cm) was used, the separation voltage was fixed, the cartridge temperature was 25 ℃, the UV detection wavelength was 214 nm, and sample was introduced from the anode with pressure injection. The mobility of neutral marker was determined with the current sudden change. The effective mobilities of the three compounds on pH were investigated, and their pKa values was determined by nonlinear simulation. Results When the ionic strength of running buffer was fixed, the pKa values of SMZ, SD and TMP were 5.69, 6.36 and 6.68, respectively; on the contrary, the pKa values of SMZ, SD and TMP were 5.71 ,6.35 and 6.60, respectively. Conclusion The proposed method is simple, fast and accurate for determination the pKa values of sulfonamides,and which isn’t affected by the ionic strength of running buffer.

    Objective To investigate effects of botong capsule on the cerebral blood flow and serum VEGF of cerebral ischemic stroke rats. Methods 70 aged SD rats were stochasticly divided into 7 groups: the normal group, model group, high dose of botong capsule group, low dose of botong capsule group, high dose of botong capsule with musk group, low dose of botong capsule with musk group, nimodipine group, then the model of cerebral ischemic stroke were established in rats by MCAO. Brain blood flow and serum VEGF were detected by using laser doppler flowmeter and ELISA. Results MCAO lowered blood flow of brain, while the high dose of botong capsule significantly increased it. In contrast, MCAO decreased the content of VEGF in blood, whereas the high dose of botong capsule remarkably raised it. Conclusion The high dose of botong capsule can improve blood flow of brain and adjust the content of serum VEGF.

    Objective To investigate the genes of aminoglycosidemodified enzymes ( AMEs ) in Stenotrophomonas maltophilia. Methods Seventeen strains of Stenotrophomonas maltophilia were clinically isolated. The AMEs (6 kinds) genes of these strains were analyzed using PCR and verified using DNA sequencing. Results The positive rate of genes of aac ( 6’ )Ⅱwas 23.5%. All of aac (3)Ⅱ,aac (3")Ⅱ ,aac ( 3")Ⅰ,aac ( 6’)Ⅰ ,aac(3)Ⅰwere negative. Conclusion The gene of aac ( 6’)Ⅱwas found in Stenotrophomonas maltophilia. The gene commonly detected in Pseudomonas increased resistance against aminoglycoside in Stenotrophomonas maltophilia.

    Objective To establish the high performance liquid chromatography(HPLC) method to determine the content of oleanolic acid(OA) and ursolic acid (UA) in Herba Glechomae for quality control of Herba Glechomae. Methods The HPLC analysis was performed in a Dionex SUMMIT high performance liquid chromatograph equipped with a PDA-100. Optimized separation conditions were as follow: reversedphase column (HypersilODS2, 5 μm, 4.6 mm×200 mm), the mobile phase of A-B-C (A, methanol; B, acetonitrile; C, 1% acetic acid) (40:40:20) with the rate of 1.0 mL•min-1, the temperature at 30 ℃, UV at 210 nm. Results The linear range for OA was 0.040.20 mg•mL-1 and for UA was 0.400.75 mg•mL-1. The intraday precision of OA and UA was 0.80% and 0.37%, individually. The recovery of OA and UA was 98.1% and 97.9%. Eleven batches of Herba Glechomae were analyzed. The content of OA in Herba Glechomae ranged from 0.17 to 0.50 mg•g-1, and UA ranged from 0.97 to 1.92 mg•g-1. Conclusion A simple, rapid and accurate HPLC method was developed for the quantification of oleanolic acid and ursolic acid in Herba Glechomae. There was high content of OA and UA in Herba Glechomae of Hubei province, which was valuable for further exploitation and application.

    Objective To investigate the effect and underlying mechanisms of the extracts of Caesalpinia sappan L (CS) on type 1 diabetes mellitus (T1DM) in NOD mice. Methods Various doses of acetoacetate extracts and nbutanol extracts of CS (ECS and BCS)were administered into 4weekold NOD mice orally to observe the effect on the onset of T1DM. All animals were killed at the age of 28 weeks, and pancreas tissues were taken for histopathologic analysis. Spleen lymphocyte proliferation was detected by MTT method and the cytokine IFN-γ and IL-10 in the culture supernatants of spleen lymphocyte was determined by ELISA analysis. Results Both extracts were shown to dosedependently decrease the incidence of T1DM significantly. Pancreatic tissues from ECS and BCStreated mice revealed a marked reduction of insulitis in the degree and severity compared with that in the control group. The proliferation of lymphocytes was decreased , while Th2 cytokine IL-10 was increased and Th1 cytokine IFN-γ was increased. Conclusion Acetoacetate extracts and nbutanol extracts of CS could confer protection to NOD mice from T1DM, and the mechanism may be related to restoring the imbalance between Th1 and Th2 cytokines in vivo.

    Objective To study the influence of different factors on the release properties of OLA from matrix tablets in vitro. Methods The sustainedrelease matrix tablets of OLA were prepared by using HPMC as the matrix materials. Factors (such as the viscosity and amount of HPMC, adhesives, concentration of moistening agent, preparation technique etc) affecting the in vitro dissolution were observed. Results Both the concentration of moistening agent and viscosity ,amount of HPMC had significant effect on the release of OLA. In contrast, the concentration of moistening agent and tablet hardness had no effect on drugrelease rate. Conclusion The tablets have desirable sustainrelease capability in vitro. The drug release in vivo will be carried on in the further test.

    Objective To establish a system of chemical pattern recognition on the quality assessment of Bupleurum scorzoneri folium Willd. Methods The content of saikosaponin a,c,d in Bupleurum scorzoneri folium Willd. Samples from 10 different areas in China was determined by RPHPLC method. The chemical data were dealed with hierarchical clustering analysis. Results The 10 samples from different habitats were classified as 3 categories according to its quality. The content of active components in Bupleurum scorzoneri folium Willd. samples from henan and sichuan was relatively higher than that from other habitats. Conclusion This method is feasible to evaluate the quality of Bupleurum scorzoneri folium Willd.

    Objective To study the stability of clarithromycin and clarithromycin entrapped in bioadhesive microspheres in low pH aqueous solutions. Methods The bioadhesive microspheres entrapping clarithromycin (Cla-Ec-Cb) were prepared using emulsification/evaporation method. The stability of clarithromycin and clarithromycin entrapped in microspheres in hydrochloric acid solution (pH=1) was determined and compared by HPLC method. Results At pH 1.0,2.0 ,3.0, the degradation halflives of clarithromycin were (5.93±0.16) min, (74.52±3.23) min, (869.58±5.82) min, respectively. Clarithromycin entrapped in microspheres was much more stable. Conclusion Clarithromycin was unstable in low pH solution. Carbopol 934p and ethylcellulose showed a protecting effect for clarithromycin from being decomposed by low pH solution.

    Objective To prepare liposomal bupivacaine and study the local anesthetic effect by injection around sciatic nerve in rats. Methods 0.5% liposomal bupivacaine was prepared with egg yolk phosphatidylcholines,cholesterol,αtocopherol and bupivacaine using film dispersion method. Its size was calculated by light microscope and electron microscope. The encapsulation efficiency was measured by ultraviolet spectrophotometer. Forty SD rats were randomly divided into 4 goups:N1(injected with saline)、N2(injected with blank liposomes)、BP(injected with 0.5% bupivacaine)、BP-L(injected with 0.5% liposomal bupivacaine). Appearance and mechanical pain threshold were evaluated after injecting around sciatic nerve, respectively. Results The morphology of liposomal bupivacaine was like sphere,its size was 150~2 000 nm,and the encapsulation efficiency was 43.6%. No local anesthetic effect was observed in ether group N1 or N2. The duration of liposomal bupivacaine(t1,t2,t3 and ttotal) was longer than bupivacaine. Conclusion Liposomal bupivacaine can be prepared successfully with egg yolk phosphatidylcholines,cholesterol,αtocopherol and bupivacaine by using film dispersion method. The duration of sciatic nerve block by bupivacaine encapsulated in liposomes can be prolonged.

    Objective To investigate the effect of curcumin on neuropathic pain in rats. Methods Thirty six male SD rats were randomly divided into 6 groups:sham operation group(Sham), solvent control group(SC),chronic constriction injury group(CCI),and curcumin treated groups(Cur30,Cur100,Cur300). Curmin30,100,300 mg•kg-1•d-1 ip.was respectively given after operation in group Cur30,Cur100,Cur300.Thermal withdrawal latency (TWL) and mechanical withdrawal threshold (MWT) of rats were measured on 1, 2 preoperative and 1, 3, 5, 7, 10, 14 postoperative days. Results TWL and MWT in CCI group were significant lower than in Sham group on every postoperative day(P<0.05,P<0.01).TWL in curcumin treated groups were higher than in CCI group on 7 postoperative day (P<0.05); TWL in group Cur100 and group Cur300 were higher than those in CCI group on 10 postoperative day (P<0.05);TWL in group Cur30 and group Cur100 were higher than in CCI group on 14 postoperative day (P<0.05);MWL in group Cur30 and group Cur100 were higher than in CCI group on 7,10,14 postoperative day (P<0.05). Conclusion Curcumin could alleviate thermal hyperalgesia and mechanical hyperalgesia induced by CCI.

    Objective To investigate the effect of controlled hypotension in functional endoscopic sinus surgery(FESS) with remifentanil at different rates. Methods Fortyfive patients(ASAⅠ~Ⅱ) were randomly divided into three groups. The patients in group R1,R2,and R3 were injected with remifentanil at a rate of 0.19, 0.28 and 0.37 μg•kg-1•min-1,respectively. Infuse propofol at a basic rate of 0.11 μg•kg-1•min-1. According to BIS value,adjusted the rate of propofol to maintain the stability of anesthesia. Made blood gas analysis and recorded MAP and HR at t0(before the operation)、t1(20 minutes after the operation)、t2(40 minutes after the opertation)and t3(after the operation). Results The MAP and HR in the patients of group R2 and group R3 were reduced at t1(P<0.05),while these in the patients of group R1 were reduce at t2. And these in the patients of group R3 continued to reduce at t2(P<0.05),which didn’t happen in group R2. The indexes of PaO2 、PaCO2 and Lac had statistic distinction only in group R3(P<0.05). Conclusion Influsion of remifentanil at a rate of 0.28 μg•kg-1•min-1 was a more suitable alternative for FESS. It could control hypotension obviously and steadily, and tissues weren’t short of oxygen at this rate.

    Objective To observe the effect of flurbiprofen axetil and tramadol on treating the recovery restlessness after general anesthesia. Methods Ninety patients were recruited into the study and divided into three groups:group Ⅰ received tramadol 0.8 mg•kg-1, group Ⅱreceived tramadol 1.6 mg•kg-1,group Ⅲ received flurbipmfen axetil 0.8 mg•kg-1 and tramadol 0.8 mg•kg-1.All the patients were intravenous administered twenty minutes before the end of surgery. The level of restlessness, pain scores and incidence of side effect were recorded. Results The level of restlessness and pain scores were significantly lower in group Ⅱ and group Ⅲ compared to those in group Ⅰ (P<0.05). No significant difference between group Ⅱ and group Ⅲ(P>0.05) was found. The incidence of nausea and vomiting in group Ⅱ was higher than that in the others. Conclusion Flurbiprofen axetil with tramadol can degrade the level of recovery restlessness after general anesthesia with less side reactions.

    Objective To optimize the water extraction and alcohol precipitation conditions of yanshuyin effervescent tablet. Methods An extraction procedure was optimized by orthogonal test. The content of chlorogenic acid which was used as the assessment index was determined by HPLC. Results The optimum water extraction process was as follows: water as extracting solvent, 1.5 h each time, 10 times solvent to the decoction pieces. The optimum alcohol precipitation process was as follows: 40 % of precipitation concentration, the alcohol deposit lasted 24 h. Conclusion The content of chlorogenic acid was used as the assessment index and determined by HPLC, which was simple, accurate, rapid, and less interference.
