    ABSTRACTTo review the progression in clinical feature, diagnosis and treatment of severe drug eruption．The domestic and foreign literatures of severe drug eruption from 1998 to 2007 were analyzed． Diagnosis of severe drug eruption depends on case history and clinical symptoms.Treatment of drug eruption is various .For treatment of severe drug eruption，it is important to diagnosis and give sufficient corticosteroid earlier．In order to control the disease and decrease the mortality rate，it is also important to strengthen supportive therapy and prevent from complications．It is an original effective method that corticosteroid hormone combined with highdose intravenous immunoglobulin treats severe drug eruption.

    (1. ABSTRACTObjectiveTo study the mechanisms of Chinese Allium fistulosum L. (PCAL) to prevent and therapy fatty liver in rats model . MethodsSixty SD rats were randomly divided into six groups: blank control group, model control group, PCAL low, middle and high groups, dongbaogantai group. Blank control group was given water and food. The other groups were fed with high fat diet, given water comprised of ethanol and treated with a small dose of carbon tetrachloride (CCl4) hypodermically once a week to establish the fatty liver model. ResultsCompared with the blank control group, the contents of TC,TG, NO, NOS, VEGF and ICAM1 in the model control group increased significantly（P＜0.01）； Compared with the model control group, the contents of TC, TG, NO, NOS, VEGF mRNA and ICAM1 in the PCAL low, middle and high groups and dongbaogantai group were decreased significantly（P＜0.01 or P＜0.05）. ConclusionPCAL has preventive and therapeutic effects on fatty liver in rats, possibly through descending blood fat and the impairments of reactive nitrogen and cytokines on liver.

    ABSTRACTObjectiveTo observe the effects of gout granula on gouty arithritis in rats at acute episode period,and analyze the mechanism of action.MethodsGout granula was used to treat the acute gouty arthritis in rat modle induced by sodium urate crystal.The control groups included blank control group, rat model group without teatment and model group treated by colchicine cream .The changes of joint circumference,the level of PGE2 in plasma and tissue,the number of leukocyte in the inflammatory exudates were detected.ResultsGout granula can reduce the joint circumference（P＜0.05，P＜0.01）,moreover,mediumdose and highdose gout granula reduce the level of PGE2 in plasma and tissue,and decrease the number of leukocytes in the inflammatory exudates（P＜0.05，P＜0.01）.ConclusionGout granula effectively inhibits the joint swelling in a dosedepended manner， possibly through PGE2 pathway and inhibiting leukocytes recruitment toward inflammation sites.

    ABSTRACTObjectiveTo study effects of aqueous extract from Herba Eclipta prostrata on S180 solid tumor and immune system of mice. MethodsThe model of S180 loading mice was established，the growth of tumor, the weight of body, spleen and thymus were investigated after intragastrical administration of aqueous extract from Herba Eclipta prostrata for 10 days. ResultsAqueous extract from Herba Eclipta prostrata at different dosage significantly inhibited tumor growth, with the ratio of 42.94%，55.91% and 52.06%（P＜0.01, all）, respectively. The thymus index of the three groups was higher than that of the normal saline group and the cyclophosphamide comparison group. ConclusionAqueous extract from Herba Eclipta prostrata shows the remarkable inhibitory effects on S180 solid tumor and enhancement on the immune system of mice.

    Effects of Melatonin on Oxidative Stress in Experimental Diabetic Nephropathy Rats

    ABSTRACTObjectiveTo investigate the influence of melatonin on the oxidative stress and the expression of nitrotyrosine（NT）as the specific symbol of peroxynitrite（ONOO）in diabetic nephropathy rats． MethodsExperimental rats were divided into normal control group（NC），diabetic nephropathy group（DN）and treatment group with melatonin（DM）. The indexes such as the body weight,the contents of blood glucose,blood cholesterol，triglyceride，malonaldehyde（MDA），superoxide dismutase（SOD）and glutathione peroxidase（GSHPx）were tested in each group after 8 weeks.The expression of nitrotyrosine（NT）was tested by immunohistochemistry method.ResultsCompared with group DN，the levels of blood cholesterol，triglyceride，MDA and NT expression were decreased significantly in group DM，the activities of SOD and GSHPx were enhanced（all P＜0.05）. ConclusionMelatonin exerts a protective effect on the kidney of diabetic rats through increasing the antioxidant capability，decreasing oxidative stress of renal tissues.

    Effccts of αGlycyrrhizic Acid on Potassium Concentration in Serum of Difference Pharmacogenetic Hartley Guinea Pig

    ABSTRACTObjectiveTo study the effects of αglycyrrhizic acid on hypokalemia in difference pharmacogenetic hartley guinea pig. MethodsIn vitro, 11βhydroxysteroid dehydrogenase was extracted from white and multicoloured hartley guinea pig kidney and mixed with inhibitory agents,such as αglycyrrhizic acid.The conversion percentage of cortisol was determined.In vive, αGlycyrrhizic acid was injected intraperitoneally during total 15 days. The activity of 11βhydroxysteroid dehydrogenase from hartley guinea pig kidney and potassium concentration in serum were measured. ResultsIn vitro,compared to multicoloured hartley guinea pig, 11βhydroxysteroid dehydrogenase in white one exhibited a lower conversion percentage of cortisol（P＜0.05）. In vive, the activity of 11βhydroxysteroid dehydrogenase from hartley guinea pig kidney and potassium concentration in serum were reduced significantly.But 150 mg•kg1 αglycyrrhizic acid, a normal dose, did not induce hypokalemia in white hartley guinea pig. ConclusionThere is significant difference of pharmacogenetics between white and multicoloured hartley guinea pig.

ABSTRACTObjectiveTo establish a high performance liquid chromatography method for the determination and pharmacokinetic study of enalapril maleate in healthy human.MethodsAgilent 1100 series HPLC system was appliedwith the column ZORBAX SBC18(4.6 mm×150 mm,5 μm).The mobile phase was composed of methanol(0.1 mol•L1)，NaH2PO4(pH=3.0)and water with a ratio of 27：20：53(V/V/V),and the flow rate was 1.0 mL•min1.The UV detection wavelength was 0－5 min 286 nm，5－8 min 210 nm and 8－10 min 286 nm.After oral administration of 20 mg enalapril maleate table in 10 voluteers，concentrationtime profile was simulated and pharmacokinetic parameters were calculated with DAS software.ResultsThe calibration curve was linear in the range from 5 to 400 μg•L1(r=0.999 9)，The main pharmacokinetic parameters after a single oral dose of 20 mg of enalapril maleate tables were as follows：Cmax =(283.03±37.50) μg•L1,tmax =(0.83±0.12) h, t1/2 =(1.57±0.32) h，AUC0t =(711.51±91.18) μg•h•L1, and AUC0∞ =(742.68±98.06) μg•h•L-1. ConclusionThe method was simple，sensitive，and accurate for clinical study of enalapril maleate.

    Effects of Tetrandrine on the Proliferation of Hypertrophic Scar Fibroblasts in Vitro

    ABSTRACTObjective To investigate the effects of tetrandrine (Tet) on the proliferation of hypertrophic scar fibroblasts in vitro. MethodsThe effects of Tet on proliferation of fibroblasts were identified by MTT assay, ［3H］TdR incorporation and flow cytometry.ERK activity was measured by immunoprecipitation. pMEK1/2、pERK1/2 and MKP1 expressions were assessed by Western blot. ResultsTet can depress the proliferation of hypertrophic scar fibroblasts and ERK activity. The pMEK1/2 and pERK1/2 expressions were inhibited，and MKP1 expression was increased by different concentrations of Tet. Conclusion The proliferation of fibroblasts is able to be depressed by Tet in the dose dependent manner through downregulating ERK signaling.

    (1. ABSTRACTObjectiveTo investigate the photoprotective effect of paisha sunscreen cream on human keratinocytes exposed in UVB radiation. MethodsAfter pretreated with paisha sunscreen cream for 24 hours, HKC cells were irradiated with different dosages of UVB. The cell viability was detected by MTT method, The supernatant was collected at 24 hours post irradiation to measure the secretion of cytokine TNFα and IL8 by ELISA. ResultsThe cell viability was inverse correlated with the irradiated dosage.Paisha sunscreen cream could boost the cell viability, reduce the secretion of TNFα. The secretion of IL8 was increased after irradiation by UVB.There was a significant difference of IL8 level between paisha sunscreen cream groups and the control group(P＜0.05).ConclusionThe damage of UVB irradiation on human keratinocyte was dosedependent. Paisha sunscreen cream played a role of photoprotection through inhibition of TNFα and IL 8 secretion and improvement of cell viability.

    (1. ABSTRACTObjectiveTo observe the antiinflammatory effects of antiacne revitalizing cream. MethodsThe antiinflammatory effects were studied in mice modle with ear edema induced by xylene and rats modle with paw edema induced by carrageenin in. ResultsAntiacne revitalizing cream obviously inhibited the ear swelling in mice , the paws swelling and the granuloma with cotton pellet in rats. ConclusionAntiacne revitalizing cream has significant antiinflammation action in animal models and shows obviously a doseeffect relationship.

    Effect of Antioxidant Vitamin C on Vascular Endothelial Function during Acute Hyperglycemia in Patients with Type 2 Diabetes

    (1. ABSTRACTObjectiveTo investigate the effect of vitamin C on endotheliumdependent arterial dilation during acute hyperglycemia in patients with type 2 diabetes. MethodsTen healthy men, 20 men with type 2 diabetes (type 2 DM) were selected. All subjects were performed oral glucose tolerant test (OGTT). Patients with type 2 diabetes were divided into 2 groups: ten patients only took OGTT ( OGTT group), and other 10 patients also accepted 2 grams of vitamin C orally (OGTT + vitamin C group). Endothelium dependent arterial dilation and endothelium independent arterial dilation were measured using high resolution ultrasound technique at the 0， 60， and 120 min during the OGTT. ResultsDuring the OGTT, the endotheliumdependent arterial dilation at 0, 60, 120 min in only OGTT group and OGTT + vitamin C group were significantly lower than those in control group, respectively (P＜0.05). Compared with 0 min, the endotheliumdependent arterial dilation at 60 min were significantly lower in only OGTT group and OGTT + vitamin C group (P＜0.05), but resumed at 120 min. Compared with only OGTT group, the endotheliumdependent arterial dilation at 60 min and 120 min were significantly increased in OGTT + vitamin C group (P＜0.05), and there was no difference between the 2 groups at 0 min. At baseline in type 2 DM patients, LDLC，Lp(a), FBG and HbA1c were negatively related to endotheliumdependent arterial dilation (P＜0.01). During OGTT, blood glucose were negatively related to endotheliumdependent arterial dilation in only OGTT group and OGTT + vitamin C group(P＜0.05). ConclusionHyperglycemia rapidly suppresses endotheliumdependent arterial dilation, indicating that prolonged and repeated postprandial hyperglycemia may play an important role in the development of diabetic angiopathy. Vitamin C improves endotheliumdependent arterial dilation during acute hyperglycemia, and this may be related to the role of vitamin C to scavenge oxygenderived free radicals.

    Comparision of Sandostatin with Homemade Somatostatin in the Treatment of Esophageal Variceal Bleeding

    ABSTRACTObjectiveTo compare the efficacy of sandostatin and homemade somatostatin in the treatment of esophageal variceal bleeding due to portal hypertension.MethodsA total of 48 patients with esophageal variceal bleeding due to hepatic cirrhosis were randomly divided into two groups.In treatment group，homemade somatostatin was infused by 0.1 mg bolus then 25 μg•h-1 continuous intravenous for 5 days.In control group，sandostatin was infused by 0.1 mg bolus then 25 μg•h-1 continuous intravenous for 5 days.ResultsHemostasis rate in 72 hours，hemostasis time，rebleeding rate，mortality rate were 83.33%,（12.0±2.3） h, 25.00%,8.33%；87.5%,（12.0±7.6） h,29.17%,4.17%，respectively in treatment group and control group.ConclusionThe efficacy of homemade somatostatin is similar to that of sandostatin in treatment of esophageal variceal bleeding due to hepatic cirrhosis.

    Preparation of the Thermoreversible Meloxicam Gel and Determination of Meloxicam Content in Gel

    ABSTRACTObjectiveTo screen an optimal formulation and the preparation process of the thermoreversible meloxicam gel and establish its determination method . MethodsOrthogonal design was used to observe the change of gelation temperature and gel strength. The content of meloxicam in thermoreversible meloxicam gel was determined by UV. ResultsThe formulation of the thermoreversible meloxicam gel was composed of P407/P188 with 15 g/15 g,7%sodium hydrate 20 mL, azone 4 mL and polyethylene glycol 400 8 g.Viscosity of the gel was proper and its stability was high. The liner response range was 1.984～19.840 mg•L-1(r=0.999 6). The mean recoveries was 98.65% (RSD 2.40%). ConclusionThe formulation is reasonable，and the preparation process was simple and easy to control with stable quality .

    Determination the Content of Naringenin in Compound Rhubarb Oral Solution by HPLC

    (1. ABSTRACTObjectiveTo explore a method to detectthe contents of naringenin in compound rhubarb oral solution. MethodsNaringenin was determined in compound rhubarb oral solution by HPLC using the chromatographic column of Hypersil ODS2（4.6 mm×250 mm, 5 μm）. The mobile phase was consisted of 0.1% acetonitrile phosphoric acid solution(21：79).Detection was performed under 283 nm wavelength and 1 mL•min1 rate. ResultsThe naringenin level was 6.19 mg•L-1 in compound rhubarb oral solution. The average recover rate of naringenin was 98.4% with 0.72% RSD . There was a good linear relationship between naringenin concentration and corresponding apices area within the range of 10－80 μg•mL1 (r=0.999 9). ConclusionHPLC was an appropriate method for quality control of rhubarb oral solution as its simple, intuitionistic characteristics and maneuverability

    Determination the Concentration of Phenobarbital in Phenobarbital Tablets by HPCE

    （1. ABSTRACTObjectiveTo establish a HPCE method to determine the concentration of phenobarbital in phenobarbital tablets. MethodsThe uncoated fusedsilica capillary(30 cm×50 μm) was used. The electrophoresis condition followed as: running voltage was 18 kV, separation temperature was 30 ℃, UV detection wavelength was 280 nm and the running buffer was 30 mmol•L1Na2HPO4 (pH=8.0) containing 75 mmol•L1 SDS. The sampler was injected by pressure for 10 s. ResultsA good linear relation ( r= 0.997 9)was observed in 8.75－200.00 μg•mL1 concentration of phenobarbital . The average recovery rate of phenobarbital was 98.47%. The RSD of intraday and interday were all less than 5%.ConclusionThe established method was simple and fast,and able to control the quality of phenobarbital tablets.

    (1. ABSTRACTObjectiveTo establish the quality standard for shengmai capsules. MethodsRadix ginseng rubra, radix ophiopogonis and fructus schisandrae chinensis in shengmai capsules were indentified by TLC. The contents of schisandrin were determined by HPLC. ResultsThe spots in the TLC were fairly clear, and no interference was shown in the blank samples. A good linear relationship was found between the aliquot amounts of schisandrin within a range of 3.992～99.800 μg•mL-1and the absorption integration values. The average rate of recovery was 99.19%(n＝6), and the RSD was 1.75%(n＝6). ConclusionThe method might be a simple, rapid and accurate analysis assaywhich could be used for quality control of shengmai capsules.

    Determination of SMZ and TMP in Compound Sulfamethoxazole Tablets by Turnover Curve Spectrophotometry

    ABSTRACTObjectiveTo determine the SMZ and TMP in compound sulfamethoxazole tablets using turnover curve spectrophotometry. Methods Compound sulfamethoxazole tablets was prepared asthe experimental solution，and，SMZ and TMP was prepared as reference solutions. Under wavelength from 210 to 220 nm, the absorbance A of the solutions was recorded every 1 nm. Then the value of A was transferred into the conversion value, and was analyzed by linear regression method. The content of the SMZ and TMP solution was calculated by the results of linear regression. ResultsAs to the conversion values, there was a good linear relationship between the sample solution and the reference solution, r=0.996 6. The average recovery of SMZ was(97.60±0.37)%，while the TMP’s was (98.70±0.28)%. ConclusionTurnover curve spectrophotometry was a quick and feasible method to determine the content of SMZ and TMP in compound sulfamethoxazole tablets.

    Determination of both Emodin and Chrysophanol Levels in Fuyanjie Suppository by HPLC

    ABSTRACTObjectiveTo establish a method for the determination of emodin and chrysophanol levels in fuyanjie suppository. MethodsThe mobile phase in HPLC was consisted of methanol0.1% and phosphoric acid (78：22)with 0.9 mL•min1 flow rate and 40 ℃ column temperature . The UV detecting wavelength was 254 nm. ResultsThe linear relationship was strong under the concentration range of 1.015－8.120 μg•mL1 for emodin and range of 1.325～10.600 μg•mL1 for chrysophanol. The average recoveries of emodin was 99.3%(RSD=1.20%), and chrysophanol was 99.3%(RSD=1.35%), respectively. Conclusion This method was proven to be simple and reliableand thus suitable for the quality control of fuyanjie suppository.

    Observation Study on Gene Detection for Hypertensive Drugs with Gene Chip Method

    ABSTRACTObjectiveTo study the gene polymorphisms of hypertension drug’s related gene in hypertension patients. MethodsTwenty patients with hypertension history from 3 to 20 years, who were treated by AT1receptor antagonism (for instance: losartan, irbesartan, valsartan),βreceptor block(for instance: metoprolol, atenolol), ACE inhibitors(for instance: enalapril, benazepril)were collected. DNA was extracted form 2 mL peripheralblood. 5 hypertension related gene sites were detected by gene chip. Results In 20 hypertensive patients, rates of wide genotype, heterozygote and mutated genotype in CYP2C9 gene site were 85%,15%and 0%,respectively； in βreceptor block site were 15%,40% and 45%； in AT1receptor site were 80%,20%and 0%；in CYP2D6 site were 25%,20% and 55%；and in hierarchize ACEI site were 40%,40% and 20%, respectively. ConclusionThe gene type detection based on gene chip might classify hypertensive patients to provide more effective therapy and lower ADR.
