    Determination of Gatifloxacin in Gatifloxacin Ophthalmic Gel by HPLC

    ObjectiveTo establish an HPLC method for the determination of gatifloxacin in the gatifloxacin ophthalmic gel. MethodsThe stationary phase was Nucleosil 120-5 C18column (4.0 mm×250 mm, 5 μm) ,the mobile phase methanol0.007 mol·L-1KH2PO4buffer (pH adjusted to 4.0 with H3PO4) (30∶70)with a flow rate of 1.0 mL·min-1. The detect wavelength 286 nm ,and the column temperature 30℃.ResultsThe calibration curve was linear within the range of 10－50 μg·mL-1, the average recovery was 99.3%(RSD＝0.63%, n＝6). ConclusionThis method is accurate and simple,it can be used for the determination of gatifloxacin in gatifloxacin ophthalmic gel .

    ObjectiveTo evaluate the costeffectiveness in the treatment of depression with three different drugs. MethodsNinety eight depression patients were randomly divided into three groups.Group A (33 cases) fluoxetine 20 mg·d-1, group B(33 cases) venlafaxine 50－200 mg·d-1, and group C (32 cases) amitriptyline 50－200 mg·d-1. All the drugs were administrated orally for 8 weeks. ResultsThe effective rates of group A,B and C were 75.76%，87.88%and 75.00%, respectively. Group B had a lower costeffectiveness ratio and higher effective rate. ConclusionVenlafaxineis superior than the other two drugs in the treatment of depression.

    ObjectiveTo study the inhibition of jinyebaidu mixture on human cytomegalovirus(HCMV) protein kinase pul97 in HCMV infection, and to study the cytopathic effect (CPE) after HCMV infection in human embryo fibroblast cells (HEL).The molecular mechanism of HCMV infection was explored. MethodsThe expression of the HCMV pul97 mRNA in infected cells was measured by semiquantitative RTPCR before and after treatment with jinyebaidu mixture or ganciclovir, and the development of cytopathic effect was observed. ResultsAfter infection for 12 hours in HEL cells, CPE developed. Both jinyebaidu mixture and ganciclovir showed obvious inhibition in the expression of HCMV pul97 mRNA. And both of them could markedly postpone the development of CPE in the infected cells. ConclusionJinyebaidu mixture could inhibit duplication of HCMV by inhibiting the expression of HCMV protein kinase pul97. The CPE caused by the HCMV infection can be postponed by jinyebaidu mixture.

    ObjectiveTo compare the effects of two cyclooxygenase-2 inhibitors，celecoxib and rofecoxib on the growth of human osteosarcoma (HOS-8603) and lung cancer cell (A-549) lines.MethodsThe anti-proliferation effects were determined by methabenzthiazuron (MTT) method. The cell growing cycle and the cell apoptosis were analyzed by flow cytometry. ResultsCyclooxygenase 2-inhibitors, celecoxib and rofecoxib had inhibitory and apoptosis-inducting effect on the growth of the two tumor cell lines. The inhibitions of celecoxib and rofecoxib to the two tumor cell lines were dose-dependent and time-dependent. The cells treated with the two drugs showed a G2/M arrest which was the peak time of apoptosis. There was a significant difference between rofecoxib and celecoxib groups (P＜0.05). And celecoxib had a stronger inhibition to HOS8603 than rofecoxib. Conclusion Cyclooxygenase2 inhibitors, celecoxib and rofecoxib have significant effect on the growth of the two tumor cell lines, and may become useful medicines in tumor treatment.

    ObjectiveTo study the regulation of the integrin β3 expression in a well differentiated endometrial adenocarcinoma cell line (Ishikawa) by estrogen (E2), progestin (P4) and heparinbinding epidermal growth factor (HBEGF). MethodsIshikawa cells were treated with nothing or with 17β-E2 (10.8 mol·L-1), P4 (10.6mol·L-1), HBEGF (10 ng·mL-1), 17β-E2 plus P4, 17β-E2 plus P4 and antiprogestin (RU 486 10.5 mol·L-1) for 48 hours. The expression of the integrinβ3 was determined with reverse transcriptionpolymerase chain reaction ( RT-PCR), western bolt analysis, in situ hybridization, and immunocytochemistry. ResultsThe expression of integrin β3 was significantly reduced by 17β-E2 treatment or by 17β-E2 plus P4 treatment. The addition of mifepristone RU486 reversed the suppression of integrin β3. P4 had no significant effect on integrin β3 expression. HBEGF significantly increased the integrin β3 expression. ConclusionThe expression of the imtegrin β3 is regulated by both steriod hormones and growth factors.

    ObjectiveTo observe the effects of traditional Chinese medicine guizhifuling (GZFL) extracture on immunity in endometriosis (EMT) rats. MethodsEndometriosis rat models were made by autograft of endometrium in abdomen. 36 rats were randomised into the control group, the model group and the test group. The test group (n＝13) were given 1 mL·(100 g)-1 of GZFL extracture (0.08 g·mL-1 ) i.g., the model group (n＝13) and the control group (n＝10) were given 1 mL·(100 g)-1 of water for 4 wk. The seroglobulin IgG, IgA, IgM in each rat were assayed by ELISA and CD＋3, CD＋4, CD＋8 and CD＋4/ CD＋8 in peripheral blood were assayed by indirect immunofluorescenece. ResultsThe seroglobulin levels of IgG, IgA, IgM in the model group were significantly higher than that of in the control group (P＜0.01). The seroglobulin levels of IgG, IgA, IgM in the test group were significantly lower than that of in the model group (P＜0.01). CD＋3, CD＋4, CD＋4/CD＋8 in the peripheral blood in the model group were significantly lower than that of in the control group, and CD＋8 in the model group were higher than in the control group. CD＋3, CD＋4, and CD＋4/ CD＋8 in the test group were higher, but CD＋8 was lower (P＜0.01). ConclusionThere were shown to that EMT may loose the immunity balance. Traditional Chinese medicine GZFL extracure can regulate the immune function, which may be one of its mechanisms how to effect the endometriosis.

    The Study of Apoptosis of MOLT4 Cells Induced by Berberine in Vitro

    ObjectiveTo investigate the effect of berberine on inducing apoptosis of human Tlymphoblastic leukemia cell line (MOLT-4).MethodsThe exponentially growing MOLT-4 cells were divided into three groups: A, B and C. Group A were given 10 or 100 mg·L-1of berberine hydrochloride, group B were given 1 mg L-1 of daunorubicin, and group C were given berberine hydrochloride plus daunorubicin. The cell survival rate were determined by MTT after 6 and 24 h incubation; the apoptosis were detected by flow cytometry, electrophoresis and microscopy.ResultsAfter incubation for 6 and 24 h, the cell survival rates in group A were 81.80% and 64.67%, group B were 98.38%and 38.46%, and group C were 68.35% and 20.51%. There was a significant difference in group C compared with group A and group B within either 6 or 24 h (P＜0.05). Berberine hydrochloride could induce the typical apoptosis changes in morphology changes and DNA ladder. The flow cytometry analysis showed that the apoptosis rates were 5.83% by 10 mg·L-1berberine hydrochloride, and 19.93% by 100 mg·L-1berberine hydrochloride at 24 h.ConclusionBerberine hydrochloride had apoptosisinducingeffect on leukemia cells and could enhance the antileukaemia activities of daunorubicin, which might provide a theoretical basis for its clinical application.

    Protective Effects of Mycelia of Glossy Ganoderma on Chemical Liver Injury in Mice

    ObjectiveTo study the effects of mycelia of glossy ganoderma on liver injury induced by carbon tetrachloride (CCl4) in mice. MethodsFifty mice were equally divided into five groups. The low, middle and high dose group were given 0.3, 0.6, 0.9 g·kg-1·d-1 mycelia of glossy ganoderma, i.g.. The control group and the model group received equivalent volumes of sodium chloride injection i.g.. After 30 d, the low, middle, high dose groups and the model group were given CCl4 by injection in mouse's abdomen. The serum levels of alanine transaminase (ALT) and aspartate transaminase (AST) were tested, and the liver pathologic histology was examined. ResultsThe serum levels of ALT and AST in the low, middle, high dose groups of was significantly decreased, and the spleen lesion and hepatic pathological injury were relieved. ConclusionThe mycelium of glossy ganoderma has protective effect on liver injury by CCl4 in mice.

    Kidneyprotect Effect of Calcium Dobesilate in Type 2 Diabetes Mellitus Rats

    ObjectiveTo study the kidneyprotect effect and possible mechanism of calcium dobesilate in type 2 diabetes mellitus rats. MethodsThe SD rats were randomized into the control group, the model group and the test group. And the type 2 diabetes mellitus rat models were produced by the use of high carbohydrate and fat food, and small dose of streptozotocin (STZ) injected. The test group were given calcium dobesilate (100 mg·kg-1·d-1) i.g.. The control group and the model group were given the same mount of water for injection. After 6 months, serum and urinary biochemistry test and kidney morphology examination were carried out. The level of endothelin (ET) in serum and the transforming growth factor β1 (TGF-β1 ) in renal cortex were determined by radioimmunoassay. ResultsThe kidney function and kidney lesions were significantly improved in the test group treated with calcium dobesilate than in the model group. The ET levels in serum and renal cortex was reduced, and the expression of TGF-β1 in test group was decreased. ConclusionCalcium dobesilate had kidneyprotect effect on the diabetic rats, which may be related to inhibition to the over expression of ET and TGF-β1.

    Synthesis and Antitumor Activities of Piperidone Farnesyltransferase Inhibitors

    ObjectiveTo synthesize the piperidone farnesyltransferase inhibitors under microwave irradiation and to observe their antitumor activities in vitro. MethodsUnder microwave irradiation condition, the target compounds were synthesized by cyclization reaction from the substance which was synthesized from 2substituted benzaldehyde by the Perkin reaction and Michael addition. The IC50 values of antitumor activities to human Hela cell and ANC-1 cell were tested by MTT method.ResultsThe target compounds under microwave irradiation were obtained within 20－45 min. The average production rate were 36.0%－67.1%. Eleven new compounds were identified by IR, 1H-NMR and ESI-MS, and all of them showed antitumor activities in preliminary experiments. IC50 values of eight compounds were below that of 5Fu. Conclusion The synthesis method of piperidone farnesyltransferase inhibitors is reliable. And piperidone farnesyltransferase inhibitors have certain antitumor activities

    Effect of Rosiglitazone on Leptin Resistant and Ovulation Induction in Women with Polycystic Ovary Syndrome

    ObjectiveTo study the leptin level in patients with polycystic ovary syndrome (PCOS) and the effects of rosiglitazone on leptin level and reproductive functions. MethodRadioimmunoassay（RIA）is used to determine the serum leptin level in fortytwo patients with polycystic ovarian syndrome and thirty controls with normal ovulation. The ovulation rate is assessed by clomifene test before and after rosiglitazone treatment. ResultsThe levels of serum leptin are significantly higher in PCOS patients than in the BMImatched controls (P＜0.01). The leptin level in PCOS patients decreased after rosiglitazone treatment, and the ovulation rate increased significantly (P＜0.01). ConclusionRosiglitazone can lower the serum leptin in PCOS patients, improve leptin resistance, and increase the ovulation rate.

    Analgesia Effect of Dexamethasone Combined with Ondansetron Compared to Tramadol in radically Operated Gastric Cancer Patients

    ObjectiveTo investigate the analgesic effects, postoperative nausea and vomiting of dexamethasone combined with ondansetron compared to tramadol in the radically operated gastric cancer patients.Methods90 radically operated gastric cancer patients were randomly dividedinto three groups (A, B, C) to do a doubleblind test: group A were given tramadol 1 000 mg, the drugs were infused in 0.9% sodium chloride injection, group B were given tramadol 1 000 mg plus ondansetron 8 mg, group C were given tramadol 1 000 mg plus dexamethasone 10 mg.The intravenous infusion of tramadol was 10 mg·h1for 10 min,and the limited dose within 4 h was 150 mg.The pain, sedation, dizziness, nausea, vomiting in the patients and the doses of tramadol and dexamethasone combined with ondansetron for 48 h were evaluated. ResultsThe pain relief in group C was the best, group A better，and group B the third. The amount of tramadol used in group B was the biggest. The dose of tramadol had no significant difference between group A and C for 48 h. Patients in group C had the least number of nausea and vomiting, and there was no significant differences between group A and B. There was no significant difference in dizziness and sedation among three groups. ConclusionOndansetron can’t lower the rate of nausea and vomiting in the radically operated gastric cancer patients, and had lower analgesia effect, requiring more tramadol. Dexamethasone can reduce the incidence of nausea and vomiting.

    Effective of Citicoline Combined with Physical Treatment on Amblyopic Patients beyond Plastic Period of Visual System

    ObjectiveTo study the effect of citicoline and physical treatment for the amblyopic beyond the plastic period of visual system. Methods54 patients were randomly divided into three groups: control group, treatment groupⅠ and treatment group Ⅱ.The control group were treated with physical therapy, the treatment group Ⅰ were treated with 500 mg citicoline, im, qd, combined with physical treatment; the treatment group Ⅱ were treated with 250 mg citicoline, im, qd, combined with physical treatment. The results were statistically analyzed. ResultsRetina light sensitivity and eyesight in treatment group Ⅰand treatment groupⅡ were improved significantly than in the control group, but there are no significant difference between treatment group Ⅰ and treatment group Ⅱ. ConclusionCiticoline combined with physical treatment have better effect than physical treatment in amblyopic patients beyond the plastic period.

    Therapeutic Effect of Jinhoujian Spray in Laryngeal Mucosal Recovery after Microwave Tissue Coagulation

    ObjectiveTo study therapeutic effect of jinhoujian spray in laryngeal mucosal recoveryafter microwave tissue coagulation (MTC). Methods76 patients with chronic laryngitis treated by MTC, were divided into two groups. 53 patients in the treatment group received jinhoujian spray after MTC, two spray each time and four times daily. 23 patients in the control group received no medicine after MTC. The laryngeal mucosal recovery in the two groups were compared 5 days later. ResultsThe total effective rate in the treatment group and in the control group were 98.1% and 82.6%, respectively. There was significant difference between two groups (P＜0.05). ConclusionJinhoujian spray has good therapeutic effect in laryngeal mucosal recovery after MTC.

    ObjectiveTo observe the effect of Ringer's solution in rapid infusion on coagulation during surgery. Methods25 patients with hysteromyoma received rapid infusion of Ringer's solution 20 mL·kg-1·h-1 during surgery. The levels of hemoglobin (Hb), platelet count (PLT), hematocrit (HCT), fibrinogen (FIB) and antithrombin Ⅲ (ATⅢ) were tested before infusion and after operation. ResultsThe levels of Hb, PLT, HCT, FIB and ATⅢdecreased significantly after operation (P＜0.05 or P＜0.01). The decrease of FIB and ATⅢconcentrations was disproportionate to the degree of hemodilution. ConclusionRapid infusion of small amount of Ringer’s solution may induce hypercoagulability during operation. The significant decrease in FIB and ATⅢis related to the hemodilution and the consumption due to thrombin formation.

    Penetration Enhancer for Acyclovir Gel

    ObjectiveTo study the percutaneous permeation of menthol and azone for acyclovir gel. MethodsDifferent five percent acyclovir gels containing different concentrations of menthol and azone were prepared. The steady state flux of acyclovir from the gel was determined in vitro on excised mouse skin using modified Franz diffusion cells. And the accumulated permeation amount and permeation rate were deterrmined. ResultsThe flux values of acyclovir by no enhancer was 0.862 mg·h-1. The flux values by 1%, 3% and 5% menthol in the gels were 0.839, 1.973 and 0.967 mg·h-1. The flux values by 1%, 3% and 5% azone in the gels were 0.693, 0.969 and 0.789 mg·h-1. The flux values by 3% menthol combined with 1%, 3% and 5% azone were 1.237, 0.997 and 0.928.ConclusionThe maximum enhancement in the acyclovir gel was 3% menthol.

    Determination of Diosyenin by Colorimetry in the Weileyi Granules

    ObjectiveTo establish a colorimetry method for the determination of diosyenin in weileyi granules. MethodsThe diosyenin was determined by ultraviolet spectrophotometry. The detecting wavelength was 406 nm. ResultsA good linearity was shown within the range of 0.002 4－0.012 0 mg·mL-1(r＝0.999 7). The average recovery was 98.37%, RSD＝2.25%(n＝5). ConclusionThis method is rapid and accurate. Its repeatability is good, and it could used to for the quality control of the weileyi granules.

    ABSTRACTObjectiveTo determine the encapsulation efficiency and the release rate of baicalin liposome. MethodsThe baicalin was separated with Sephadex G50, and determined by ultraviolet spectrophotometry. ResultsBaicalin liposome had a good linearity within the range of 4－20 μg·mL-1 (r＝0.999 4, n＝6), the average recovery was 98.83%, RSD＝2.26%.ConclusionThis method is simple, accurate which can be used for the determination of encapsulation efficiency and release rate of baicalin liposome.
