    ObjectiveTo screen the appropriate macroporous resins and the optimal technique for the enrichment of total polyphenols from Rosa rugosa Thunb . MethodsStatic and dynamic adsorptiondesorption methods were adopted, and the optimal conditions were evaluated by two equivalent important factors: the content of the total polyphenols and the weight of the effective fraction to determine the best technology. ResultsThe HPD600 macroporous resin had the best enrichment efficiency with the sample concentration: 5.50 mg•mL1; elution solvent concentration: 50% (V/V) ethanol; elution volume: 5 BV (resin bed volume); elution speed: 4 BV•h1. The total polyphenols concentration was over 60% (m/m) by enriching. ConclusionHPD600 is effective to enrich the total polyphenols from Rosa rugosa Thunb. This technology is simple and suitable for industrialization.

    ObjectiveTo investigate the pyrogens removing technology with activated carbon and ultrafiltration by quantitative detection of bacterial endotoxins. MethodsThe content of bacterial endotoxins was detected by keniticturbidimetry and the removal efficacy was studied before and after using different concentrations of active carbon and ultrafiltration membranes at different cut off molecular weight in medicine solution. ResultsThe removal efficacy of bacterial endotoxins was 62.21%, 76.21%, 81.03% by using three concentrations of active carbon at 0.03%,0.10%,0.30%, respectively in Panax Notoginseng Saponins solution, and it was above 91% with ultrafiltration membrane at cut off molecular weight both 50 000 and 100 000. ConclusionThe quantitative relationship between activated carbon and their bacterial endotoxins removal efficiency and which by ultrafiltration are clarified in our study. It provides evidences for removing pyrogens from injections in production technology.

    ObjectiveTo study the applicability of ultrafiltration of Paeonol solubilized by tween80 and hydroxypropylβcyclodextrin. MethodsTemperature, pressure, concentration and membrane pore size were selected as factors for orthogonal design method. The concentration was determined in medical solution before and after ultrafiltration, and the transmittance rate(TS) was calculated to observe the content variation of Paeonol solubilized by tween80 and HPβCD. ResultsConcentration had markedly influence on TS of tween80(P<0.01); the four factors had no significantly impact on TS of, HPβCD,but TS of Paeonol solubilized by tween80 was lower. ConclusionThe applicability of ultrafiltration of unsoluble active components dissolved by HPβCD is good, while tween80 is not suitable for assisting ultrafiltration of less soluble components.

    ObjectiveTo investigate the therapeutic effect and mechanism of thalidomide(Thd) on type Ⅱ collagen induced arthritis of rats. Methods30 female SD rats were randomly divided into control, model and Thd treatment groups. CIA model was induced by type II collagen. Thd group was treated with Thd 150 mg•kg1 the day after model establishment, and the control and model groups were administered with normal saline 15 mL•kg1,for 6 weeks. Severity of joint inflammation was scored during the treatment and right metapedes were scored by radiology technique posttreatment. The microvessel density(MVD) of synovium was detected by immunohistochemistry method, the active expression of VEGF and matrix metalloproteinase1, 2, 3, 9 in serum were tested by Western Blot assy. ResultsThe active expression of VEGF and MMP1, 2, 3, 9 in blood vessels of model group obviously increased compared with the normal control group; meanwhile, the inflammatory arthropathy index, radiological and synovium histopathology changes in thalidomidetreated rats were all conspicuously decreased compared with those in the model group (P<0.05 or P<0.01). ConclusionThalidomide can remarkably improve arthritis symptoms and radiographic changes of CIA rats. It could inhibit synovium angiogenesis by downregulating expression of VEGF and MMP1, 2, 3, 9.

    ObjectiveTo study the antidiarrheal and in vitro antibacterial effects of jiawei houpu wenzhong decoction. MethodsThe conventional mouse model of diarrhea was used to carry out the antidiarrheal experiment; the minimal inhibitory concentration(MIC) and minimal bactericidal concentration (MBC) were determined for decoction by two fold tube dilution test and cupplate method, respectively.ResultsJiawei houpu wenzhong decoction reduced the number and rate of watery feces induced by rhubarb(P<0.05,P<0.01), The MIC of kiawei houpu wenzhong decoction against escherichia coli,staphyloccocus aureus rosenbach,bacillus proteus, shigella dysenteriae, salmonellatyphi were 62.25,15.56,62.25,31.13,31.13 mg•mL1, respectively;and MBC of which against escherichia coli,staphyloccocus aureus rosenbach,bacillus proteus, shigella dysenteriae,salmonellatyphi were 62.25,31.13,62.25,62.25,31.13 mg•mL1 respectively. ConclusionJiawei houpu wenzhong decoction has antidiarrheal and in vitro antibacterial effects.

    ObjectiveTo study the antiradiation effects produced by 17βestradiol3 allyl ether (ES1) and 17βestradiol3isopentenyl ether(ES2).MethodsUsing ethinylestradiol and rubidate as positive control, the mice receive 6.5 Gy yrays emitted from the 137Cs radiation resource. The number of white blood cell(WBC), the number of nucleated cells in bone marrow(BMNC), DNA content, colony forming unitspleen (CFUS), spleen index and thymus index were detected to evaluate the protection of ES1 and ES2 to the mice against radiation.ResultsCompared with the control group, the three dosage groups of both ES1 and ES2 exhibit better antiradiation effects, including the aspects of WBC,BMNC,DNA content, CFUS, spleen index and thymus index (P<0.05, P<0.01), especially,the improvement of the WBC (P<0.01). Therefore, we can conclude that the two compounds can protect the hematopoietic function and promote the immunity of the mice. ConclusionES1, ES2 have the effects of antiradiation and may become an oral antiradiation protector with higher efficiency, lower toxicity.

    ObjectiveTo investigate the protective effects and mechanisms of ACEI (captopril),Milkvetch root and their combination on human umbilical vascular endothelial cells (HUVECs) damage induced by hydrogen peroxide(H2O2). MethodsHUVEC were injuried for 4 hours by 2.00 mmol•L1 H2O2 after they were incubated with different dosages of captopril, Milkvetch root and their combination for 24 hours. The morphology change of apoptosis was observed by AO/EB fluorescent staining. Apoptosis ratio of HUVEC was detected by FCM. Expression of Bcl2 protein was detected by Western blot technique. ResultsThe early cell apoptosis ratio in the normal control and (captoprilL, M, H/Milkvetch rootM, H) were significantly lower than those in the H2O2 oxidative control (P<0.05 or P<0.01). The total cell apoptosis ratio in the normal control and (captopril/Milkvetch rootM, H)+ H2O2 groups were significantly lower than those in the H2O2 oxidative control(P<0.05 or P<0.01) except captopril/Milkvetch rootL+H2O2 group. Besides, the content of important inhibition apoptosis protein Bcl2 of cell in the normal control and (captopril/Milkvetch rootM, H)+ H2O2 groups were significantly higher than those in the H2O2 oxidative control (P<0.05 or P<0.01). Significant differences were found between drug combination groups and single use groups in early apoptosis rate, total apoptosis rate and Bcl2 content (P<0.05 or P<0.01). ConclusionCaptopril, Milkvetch root and their combination can decrease the apoptosis ratio induced by H2O2, increase the level of Bcl2 in a dose dependent way. The combined use of captopril and Milkvetch root injection is better than the single use of captopril and Milkvetch root.

    ObjectiveTo investigate effects of puerarin on urinary albumin excretion rate of diabetic rats, and explore the protection of which against renal damage. MethodsSD rats were randomly assigned to normal control group (A) , diabetic group (B),puerarin at a lowdose(C), middledose (D), highdose group (E). Rats in group C,D,E were i.p. treated with puerarin 40,80,160 mg•kg1•d1, respectively; those in group A and B were treated with sodium chloride at the same volume. The urinary excretion rates of albumin were tested at the second, fourth and eighth week. ResultsThe urinary protein excretion of albumin in group C,D,E at three time points were all higher than those in group A, but much lower than those in group B (P<0.01 or P<0.05). ConclusionPuerarin could attenuate glomeruli injury, reduce urinary albumin excretion of diabetic rats and play a role in protecting kidneys of diabetic rats.

    ObjectiveTo study the effects of Paclitaxel on immune function of human monocyte derived dendritic cells. MethodsMonocytes were isolated and fed with fetal bovine serum, granulocytemacrophage colonystimulating factor (rhGM CSF) and interleukin4 (rhIL4), paclitaxel or wihtout paclitaxel, to prepare mature DC(mDC). The DCs immunophenotypic(CD1a、CD86、HLADR) expression, endocytic activity and apoptosis were measured by flow cytometry. Allogeneic T cell activation by DCs was tested by mixed lymphocyte reaction (MLR). Cytokines secretion of IL10,IL12,TNFα were detected by ELISA. ResultsThe level of CD86、CD1a、HLADR expression on DC in paclitaxel treated group was lower than those in the control group ( P<0.01); paclitaxel inhibited the endocytic and Tcell stimulating activity of DCs;induced DC apoptosis ( P<0.01);and reduced the secretion of IL10、IL12 and TNFα of DC ( P<0.01).ConclusionPaclitaxel can inhibit immune function of human monocyte derived dendritic cells, which could be one of the mechanisms of preventing restenosis post stent implantation.

    ObjectiveTo investigate the effects of paeoniflorin on serum levels of TNFα、IL6、IL10 in rat hepatic fibrosis induced by carbon tetrachloride. MethodsSixty SpragueDawley rats were divided into 6 groups: normal control group (A),model group (B),paeoniflorin at a low dose (C1),medium (C2),high (C3) and silymarin contrast group (D). All rats except group A, were subcutaneously injected with carbon tetrachloride (3 mL•kg1 body weight ) twice a week for 8 weeks. After establishment of the model, rats in group C1,C2,C3 were administered with paeoniflorin at the doses of 5 g•kg1 body weight (C3),2.5(C2),1.2 g•kg1 (C1) for 8 weeks, those in group D were treated with silymarin at a dose of 50 mg•kg1 body weight. The changes of serum levels of TNFα、IL6 、IL10 were measured by ELISA. ResultsCompared with group B,the serum levels of TNFα and IL6 in group A were significantly decreased( P<0.01), those in group C1,C2,C3 and D reduced as well ( P<0.05); while IL10 were obviously elevated in group A(P<0.01) and group C1,C2, C3 and D(P<0.05). ConclusionPaeoniflorin could reduce hepatic fibrosis in rats by lowering TNFα、IL6 and raising IL10 dosedependently.

    ObjectiveTo study the GAD related protective effect of tetramethlpyrazine (TMP) on the respiratory inhibition induced by hypoxia in rats brainstem. MethodsThe immunohistochemical technique were used to investigate the effect of hypoxia on the GAD expression in the brainstem and the antagnistic action of TMP. ResultsIn the brainstem, GAD positive neurons were mainly existed in the HN, LRN, ambiguous nucleus (Amb), preBotzinger complex (PBC), FN and TZ. In contrast to the air control and TMP control groups, the level of GAD immunoreactivity was enhanced remarkably by hypoxia. In the rats with hypoxia plus TMP, the positive expression of GAD was obviously weaker than that in the rats with hypoxia alone (P<0.05). ConclusionTMP has protective effects on the respiratory inhibition induced by hypoxia,and GAD may be involved in the process.

    ObjectiveTo study bioequivalence of domestic emtricitabine capsules and tablets in healthy human subjects,by using imported emtricitabine capsules as reference. MethodsIn a 3preparation and 3period optimized latin square design, 24 healthy male volunteers were given a single oral dose of 200 mg emtricitabine as capsule (A), tablet (B) and reference (R). The concentration of emtricitabine in plasma was determined by HPLC. The pharmacokinetic parameters were processed by DAS program for statistic analysis. ResultsThe pharmacokinetic parameters of A,B and R were as follows: Cmax (2.28±0.63), (2.31±0.57) and (2.28±0.70) mg•L1; tmax (1.02±0.34), (1.10±0.31) and (1.03±0.33) h;AUC016h (8.50±1.73), (8.47±1.60 ) and (8.20±2.52) mg•L1•h;AUC0∞ (8.82±1.76 ), (8.78±1.63 ) and (8.56±2.54) mg•L1•h;t1/2 (3.20±0.50), (3.28±0.65) and (3.26±0.87) h. The relative bioavailability of the test preparations was (100.7±11.4)%(A) and (97.3±26.5)%(B). ConclusionThe domestic emtricitabine capsules and tablets are bioequivalent to the imported emtricitabine capsules.

    ObjectiveTo investigate the effect and adverse reaction of combination therapy of ultrashort wave and Tylox on periarthritis of shoulder. Methods140 outpatients of periarthritis of shoulder were randomly divided into treatment group(75) and control group(65). Patients in both groups were received ultrashort wave 30 min,qd, those in the treatment group were treated with Tylox one tablet, tid, for consecutive 7 d, additionally. Visual analogue scale and range of motion were observed to evaluate the curative effect. Adverse reaction was studied at the same time. ResultsAfter one week of treatment, the pain of both groups was ameliorated, and the treatment group was superior to the control (P<0.05). The motion of shoulder joint was improved in both groups as well (P<0.05). Only a few patients in the treatment group had adverse reactions, including dizziness, lethargy, nausea, vomiting, constipation.ConclusionTylox cotreated with ultrashort wave could relive pain caused by periarthritis of shoulder and improve the range of motion of shoulder with slight adverse reaction.

    ObjectiveTo prepare a sunscreen ointment special for PLA soldiers by using the total flavones in falcate crazyweed herb. MethodsThe sunscreen emulsion was prepared by using the oil in water emulsion formulation, added with flavonoids ingredient extracted and refined from falcate crazyweed herb. The quality was evaluated and its content was determined by ultraviolet spectrophotometry. ResultsThe sunscreen emulsion is stable, uniformity,easy to squash and has a good effect on absorbing ultraviolet radiation and high sunproof factor. ConclusionThe cream accorded with the standards on the PRC QBSkin care cream (QB/T18572004), and it’s hopeful to to be developed a new natural sunscreen cream.

    ObjectiveTo optimize the water extracting technology of buxue granule. MethodsAn orthogonal test was adopted in the study. The harvesting rate of the extract,contents of ferulic acid were detected.Data processing was carried out with the multiple guidelines grading method for optimizing the extraction condition.ResultsThe optimized extraction technology for buxue granule was shown to be using 10 times the volume of water,decocting for two times and 1h in each time. ConclusionThe optimized extraction is rational and efficient.

    ObjectiveTo establish a method for trace elements determination of Vernonia Anthelmintica Willd. MethodsCu、Zn、Fe、Ca、Mg、Mn、Sr in Vernonia Anthelmintica Willd was detected by inductively coupled plasma atomic emission spectrum(ICPAES) simultaneously after HNO3H2O2 digestion. ResultsThe RSD(n=9) was 0.78%~3.01%, while the recovery of the method for every trace element in Vernonia Anthelmintica Willd was 94.67%~112.70%, the results obtained were satisfactory. ConclusionThis method is brief, rapid and accurate, which can be used to analyze and measure the trace elements of Vernonia Anthelmintica Willd. It is found that Cu、Zn、Fe、Ca、Mg、Mn、Sr are pretty rich in the Vernonia Anthelmintica Willd, which is valuable for the further study of the pharmacological action of Vernonia Anthelmintica Willd.
