ObjectiveTo investigate the therapeutic effects of allitridin on organs pathological changes of systemically disseminated MCMV infected mice. MethodsAfter inoculation with Smith strain of MCMV, sixty BALB/c mice were randomly divided into allitridintreated and placebotreated groups. The therapy started at 24 hours after the inoculation. Allitridin was given to mice via intraperitoneal (i.p.) route once a day at 25 mg·kg1·d1;while the same volume of 0.9% sodium chloride solution was given in placebotreated group, and both treatments lasted for 120 d. 3 mice from each group were sacrificed at 1, 3, 7, 14, 28, 45, 60, 75, 90 and 120 d respectively after therapy. The liver, lung, kidney, and myocardium were fixed, sliced at a thickness of 5 μm and stained with hematoxylin and eosin (HE) for histology examination. ResultsThe pathological score of liver, lung and myocardium from allitridintreated group reached peak level on day 7, and kidney on day 14, then all descended gradually. Compared with placebotreated group, the pathological injury was ameliorated in allitridintreated group: the histological activity index (HAI) of livers from both groups were obviously different on day 14 (P<0.01);the Gloor score of lungs were significantly different since day 28(P<0.01);the difference of the pathological score of kidneys and myocardiums was significant since day 14(P<0.05), and became more predominant after day 90(P<0.01). Moreover, on day 120, the liver and lung of allitridintreated mice were nearly normal, while the kidney and myocardium still had some degree of pathological injury. ConclusionAfter treated with allitridin for 2 weeks, the pathological injuries in liver, lung, kidney and myocardium of infected mice were ameliorated obviously, especially in liver and lung.
