    To investigate the effect of traditional Chinese medicine jinyebaidu on early pregnant guinea pigs infected with guinea pig cytomegalovirus (GPCMV) and its safety in pregnancy.Methods:①A total of 75 pregnant guinea pigs in the first trimester were randomly divided into three groups: blank control group were treated with nothing special; positive control group were only inoculated with 1ml suspension of GPCMV intraperioneally; treatment group were given jinyebaidu through stomach perfusing after being inoculated. The guinea pigs were killed after 20 days and the GPCMV infection rate was examined in the blood and organs ②90 pregnant mice were randomly divided into three groups: blank control group were given nothing; positive control group were treated with vitamin A and treatment group accepted jinyebaidu. The effect on pregnant mice and the development of their offsprings were followed up. Results:①the GPCMV DNA positive rate of blood in the treatment group was lower than that of positive group(χ2＝39.4, P＜0.01) significantly. The rates of viremia manifestation and death were 96.0%(24/25) and 4.0% (1/25) respectively in the positive group, but no infection sign was seen in the treatment group. ②Jinyebaidu has no effect on the offsprings.Conclusion:Traditional Chinese medicine jinyebaidu markedly reduces the GPCMV maternal infection rate. The medicine is safe when used in pregnant mice.

    To investigate the influence of radix isatidis on the lipid peroxidation (LPO) level and superoxide dismutase (SOD) activity in the serum of endotoxin (ET)induced disseminated intravascular coagulation (DIC) rabbits. Method:Experimental models were duplicated by intravenous injection of ET twice. The levels of LPO and SOD in DIC rabbits serum were determined with thiobarbituric acid colorimetric method and xanthine oxidase method respectively. Results:Radix isatidis and D fraction from radix isatidis could apparently reduce LPO level and increase SOD activity in the serum of ETinduced DIC rabbits. Conclusion:D fraction from radix isatidis which can inhibit biological activities evoked by ET has the effect of antilipid peroxidation and scavenging oxygen free radicals, and constitutes an important substantial basis by which radix isatidis exerts the antiendotoxin activities.

    The Protective Effect of Garlicin on Lipid Peroxidization Injury in Ulcerative Colitis of Rat

    To observe the effects of garlicin on lipid peroxidation in ulcerative colitis of rats. Methods:The ulcerative colitis models of rats were induced with trinitrobenzenesulfonic acid(TNBS). The rats were randomly divided into 4 groups:① normal group; ②TNBS＋NS group; ③TNBS＋GarL group; ④TNBS＋GarH group. The activity of SOD and the content of MDA in all rats were evaluated the macroscopical and histological changes of the colon were observed. Results:In TNBS＋NS group and TNBS＋GarL group, the activity of SOD was lower and content of MDA was higher than those in normal group (P＜0.01). In TNBS＋GarH group, SOD activity was higher and MDA content was lower than those in TNBS＋NS group and TNBS＋GarL group (P＜0.01). The macroscopical and histological changes were slighter than those in TNBS＋NS group and TNBS＋GarL group.Conclusion:Garlicin has protective effects on ulcerative colitis of rat by inhibiting lipid peroxidation.

    Determining the Activity of CYP3A in Human Liver Microsomes by HPLC

    To express CYP3A activity of human liver microsomes by the ratio of Ndepropylpropafenone to propafenone. Methods:In 1 g·L1 microsomal protein propafenone was incubated at 37℃ for 1 hour. Ndepropylpropafenone and propafenone were determined by HPLC. Results:The linear regress equations for propafenone and Ndepropylpropafenone were Y＝0.452 0X＋0.003 029(r＝0.999 7) and Y＝1.137 3X＋0.012 740(r＝0.998 5). Conclusion:The maximal ratio of Ndepropylpropafenone to propafenone was 1.54, which shows that CYP3A has a characteristic of saturation.

    The Effects of Amiloride on Proliferation of Hepatic Stellate Cells of Rat

    To investigate the effect of amiloride on proliferation of hepatic stellate cells(HSC) of rat. Method:HSCT6 were cultured in different concentration of amiloride for 24 hours. MTT colorimetry was used for assaying proliferation of HSCT6. Cytotoxicity was measured by LDH colorimetry. Results:＞1.875 μmol·L1 amiloride can inhibit HSC proliferation. Conclusion:Amiloride can inhibite HSC proliferation and the effect is not due to nonspecific cytotoxicity of amiloride.

    Effect of Rhubarb on Endotoxin in a Rat Model of Early Lung Injury Caused by Gut Derived Infection

    To observe the change of endotoxin in gut derived lung injury treated by rhubarb. Method:The cecal ligation and perforation(CLP) was utilized to produce abdominal infection in rats and rhubarb was poured into the stomaches of anesthetized rats once every day. Six groups were sacrificed respectively before operation and 24，48，72，96 and 120 hours after operation. The pulmonary vascular permeability and the neutrophil percentage of bronchoalveolar lavage fluid (BALF) were detected. The concentrations of endotoxin in plasma, lung and BALF were assayed. Results:The pulmonary vascular permeability, the neutrophil percentage of BALF, and the concentrations of endotoxin in plasma, lung and BALF increased progressively, but they increased more slowly and less in degree in the treatmentgroup than in the infectiongroup. Conclusion:Rhubarb can significantly modify lung injury through preventing increase of endotoxin and neutrophils in lung tissue.

    To establish a model of collagen induced arthritis(CIA) in rats and investigate the effect of leflunomide and methotrexate on CIA in infancy rats.Methods:We reproduced an experimental arthritis model on healthy wistar rats ［female, 4 to 5 weeks old, (150±10) g weight］ by intradermal injection of Ⅱ type collagen. After that, all rats were divided into three groups randomly and treated differently: In Group LEF, the rats were treated with LEF＋Pred for 4 weeks; in Group MTX, the rats were treated with MTX＋Pred for 4 weeks; in Group blank control, rats were treated with placebo for 4 weeks. Then we examined arthritis index, ankle joint volume, balance of movement, the Xray of the rats' ankle, the histopathology of ankle joint, liver and kidney, the serum IgG and Creactive protein. Results:The arthritis index, ankle joint volume, balance of movement, the Xray of ankle joint and the histopathology of ankle joint showed more significant amelioration in Group LEF and Group MTX than the control group, P＜0.05. There was only one rat appearing hyaline degeneration in Group LEF and 6 rats appearing hyaline degeneration and spotting necrosis in Group MTX. There was significant difference in the pathologic results of hepatic tissue between these two groups(P＜0.05). Conclusion:LEF and MTX both can inhibit the swollen degree of the rats' ankles and synovium inflammatory responses and improve the motive function, but LEF has less side effects than MTX.

    The Effect and Safety of Treatment with a Combination of Spironolactone and ACEI for Patients with Heart Failure

    To investigate longterm effect and safety of treatment with spironolactone and ACEI for patients with heart failure.Methods:12 patients who had been administrated with digoxin and urinative before the trial were added with spironolactone and ACEI. Doses of spironolactone were 20～60 mg·d1. The use of ACEI: 2 patients with enalapril, 3 patients fosinopril and 7 patients perindopril. The patients were followed up for 2 to 6 years. Results:The function of heart of 3 patients recovered from NYHA grade3 to grade2, 5 patients from grade4 to grade3(4 cases) and to grade2(1 cases), 3 patients reached grade1. The ratio of heart and chest in 4 patients were decreased. The serum potassium of 12 patients was 4.03～5.01 mmol·L1 with an average of (4.60±0.32) mmol·L1, 2 patients suffered from diarrhea, 1 male patient had breast growth in the observation. Conclusion:The longterm combined treatment with spironolactone and ACEI for patients with heart failure is effective and safe, if the method of therapy is reasonable.

    Effects of Perindopril, Metoprolol and Amlodipine on Vascular Endothelial Function in Patients with Essential Hypertension

    To study the effects of perindopril metoprolol and amlodipine on levels of plasma nitric oxide (NO), calcitonin generelated peptide (CGRP) and endothelin (ET) in patients with essential hypertension(EH). Method:112 patients with EH were divided into three groups:perindopril group 37, metoprolol group 38 and amlodipine group 37. The drug for each group is administered in 3 different doses, each dose lasting for 2 weeks. The increase of dosage is on the basis of poor effect of lowering the blood pressure: perindopril for 2 mg→4 mg→8 mg, qd; metoprolol form 12.5 mg→25 mg→50 mg, bid; amlodipine from 2.5 mg→5 mg→10 mg,qd. A course of treatment is 3 months for every group. The plasma N0, CGRP and ET levels were observed before and after the treatment and compared with the health control group. Results:The levels of NO,CGRP in patients with EH were lower than those in the normal group before treatment (P＜0.05), and the level of ET was higher (P＜0.05); the levels of NO,CGRP after treatment were higher and the levels of ET were lower after treatment than those before treatment in perindopril group (P＜0.05); the levels of CGRP after treatment were higher and the levels of ET were lower after treatment than those before treatment in amlodipine group (P＜0.05); there were no changes of NO, CGRP and ET levels after treatment in metoprolol group. Conclusion:Perindopril and amlodipine have the action of improving vascular endothelial function deterioration which is often present in patients with EH and metoprolol has no this effect.

    A Comparison of Effects of Ginkgo Biloba Flavonal Glyco and Compound Salvia Miltiorrhiza Tablets in Treating Patients with Angina

    To compare the therapeutic effects of ginkgo biloba flavonal glyco and compound salvia miltiorrhiza tablets in treating patients with angina. Method:68 patients were divided randomly into study group(n＝38, ginkgo biloba flavonal glyco, 2 tablets, tid, po) and control group(n＝30, compound salvia miltiorrhiza, 3 tablets, tid, po). Before and 4 weeks after the treatment, the clinical symptoms, electrocardiograms, cardiac functions, hemorheology, dosage of nitroglycerin and the adverse effects were observed.Results:The study group had better therapeutic effects in improving clinical symptoms(P＜0.05), electrocardiograms, cardiac functions and hemorheology(P＜0.05); and needed a smaller dosage of nitroglycerin than the control group(P＜0.05). There were no obvious adverse effects in both groups.Conclusion:Ginkgo biloba flavonal glyco is safe and efficient in the therapy of patients with angina and it is superior to compound salvia miltiorrhiza.

    To prospectively assess the magnitude of changes in the arterialtoend tidal carbon dioxide gradient ［P(aET)CO2］ as well as in the ratio of physiological dead space to tidal volume (Vdphys/Vt) during controlled sodium nitroprussideinduced hypotensive anaesthesia, and to evaluate whether or not ventilatory requirements remain unaltered during this procedure. Method:Twenty adult patients with American Society of Anesthesiologists' physical status I and II undergoing nasal sinus surgery were selected. A standard anaesthetic procedure was followed for all cases, using propofol, succinylcholine, fentanyl, and isoflurane. Mean arterial blood pressure (MAP) was reduced to 50%－60% of baseline in all patients using a sodium nitroprusside infusion. The endtidal carbon dioxide tension(PETCO2), the arterial carbon dioxide tension(PaCO2), MAP, heart rate(HR), and expiratory minute volume were recorded during a period of normal arterial blood pressure(T0), during hypotension (T1, T2)and during a period of normal arterial blood pressure after hypotension(T3). Results:A significant decrease in PaCO2 and PETCO2 from T0 to T1 and T2 (P＜0.01) was noted, as was a significant increase in P(aET)CO2 and in the Vdphys/Vt ratio (P＜0.01) during the same period. Conclusion:The decrease in PETCO2 does not reflect the changes in PaCO2. The larger increase in P(aET)CO2 is mainly due to the increase in the Vdphys/Vt ratio. During anaesthesia, once normocapnia is achieved with normal arterial blood pressure, there is hardly any need to change the ventilation during the period of controlled hypotension.

    Study on Quality Standard of Zhitonghewei Granules

    To establish a method for quality control of zhitonghewei granules.Method:Rhizoma corydalis, pericarpium citri reticulatae and fructus crataegi were indentified by TLC. A method of thinlayer UV spectrophotometry was used to determine the content of tetrahydropalmatine. Results:The study on the quality control showed that the characteristics for identification by TLC were distinct. The quantification method of the tetrahydropalmatine had the linear range of 5～30 μg·mL1, r＝0.999 6. The average recovery was 96.5% and it's RSD was 1.97%. Conclusion:The methods for identification and quantification are convenient and accurate, being suitable for qualify control of zhitonghewei granules.

    Simultaneous Determination of Both Ferulic Acid and Tetramethylpyrazine in Serum by Reverse Phase HPLC

    To establish a method for determination of ferulic acid (FA) and tetramethylpyrazine (TMP) in serum sample with boiling waterbath. Method: On C18ODS column (4.6 mm×250 mm, 5 μm), use methanolwater(containing 1% acetic acid)＝7∶18 as the mobile phase and make UV detection at 280 nm. Results:The method of FA was liner within the ranger of 30.2～70.8 μg·mL1, Y＝413.12X＋57.51, r＝0.997 4; the method of TMP was liner within the ranger of 10.1～50.7 μg·mL1，Y＝308.64X＋41.32, r＝0.992 2。 Conclusion:Compared with traditional extraction, this method is simple, rapid and sensitive, with the advantage of not carrying in any impurities.

    To establish HPLC method for the determination of bilirubin in rengongniuhuang and its preparation. Method:Bilirubin in rengongniuhuang and its preparation was assayed by RPHPLC with a MICROSORBMV C18 column (4.6 mm×150 mm, 5 μm). Methyl acohol tetrahydrofuran0.5% acetic acid (80∶10∶10) was used as the mobile phase. The flow rate was 1.0 mL·min1. The detection wavelength was 450 nm. Results:The linear range of bilirubin was 0.12～1.20 μg(r＝0.999 7). The average recovery(n＝3) was 97.64% (RSD＝0.64%). Conclusion:The method is rapid, simple and accurate.

    To investigate the economical way and effectiveness in different methods with Ticarcillin/clavulanate potassium and Amaxicillin/clavulanate potassium in the treatment of lower respiratory infection.Method:Pharmacological economic principle was adopted to analyze the costeffectiveness of different therapy methods. Results:The results showed that the two different therapy methods were the same in effectiveness. But the cost of sequential therapy was lower. Conclusion:Sequential therapy is the best method to be chosen for the treatment of lower respiratory infection.

    Clinical Analysis to 40 Disulfiramlike Reactions Caused by Cephalosporin

    Make clinical physicians pay great attention to disulfiramlike reactions caused by cephalosporin. Method:Backwardanalyze 40 disulfiramlike reactions caused by cephalosporin between January to June of 2003 in union hospital.Results:① Disulfiramlike reactions can be seen in different sexes, ages, primary diseases. ②Disulfiramlike reaction can be caused by cephazolin, cefoperazone, cefoperazone and sulbactam, latamoxef and so on. ③Symptoms are flush, chest distress, headache, palpitation, nausea, vomit, shortbreath, dysphoria, hypotension, angina, dyspnea and so on. ④Besides drink all kinds of wines, repeating skin disinfect by ethanol or take preparations containing ethanol such as huoxiang zhengqishui can also cause disulfiramlike reactions. ⑤Disulfiramlike reaction appears when cephalosporin was being used or within 72 hours after cephalosporin had been used. ⑥Disfiramlike reaction appears 1040 minutes after drink. ⑦Disulfiramlike reactions are normally be relieved in 12 hours after treatment. Conclusion:Physicians and nurses should pay great attention to disulfiramlike reactions caused by cephalosporin, especially to the old and those patient with cardiovascular diseases.
