To establish a model of collagen induced arthritis(CIA) in rats and investigate the effect of leflunomide and methotrexate on CIA in infancy rats.Methods:We reproduced an experimental arthritis model on healthy wistar rats [female, 4 to 5 weeks old, (150±10) g weight] by intradermal injection of Ⅱ type collagen. After that, all rats were divided into three groups randomly and treated differently: In Group LEF, the rats were treated with LEF+Pred for 4 weeks; in Group MTX, the rats were treated with MTX+Pred for 4 weeks; in Group blank control, rats were treated with placebo for 4 weeks. Then we examined arthritis index, ankle joint volume, balance of movement, the Xray of the rats' ankle, the histopathology of ankle joint, liver and kidney, the serum IgG and Creactive protein. Results:The arthritis index, ankle joint volume, balance of movement, the Xray of ankle joint and the histopathology of ankle joint showed more significant amelioration in Group LEF and Group MTX than the control group, P<0.05. There was only one rat appearing hyaline degeneration in Group LEF and 6 rats appearing hyaline degeneration and spotting necrosis in Group MTX. There was significant difference in the pathologic results of hepatic tissue between these two groups(P<0.05). Conclusion:LEF and MTX both can inhibit the swollen degree of the rats' ankles and synovium inflammatory responses and improve the motive function, but LEF has less side effects than MTX.
