    Objective To study the effect of ranolazine on the contents of triglyeride (TG), fatty acid translocase (FAT)/CD36 and insulin secretion in insulinoma cell line NIT-1 induced by high glucose and saturated fatty acid. Methods NIT-1 cells were cultured with high glucose, saturated fatty acid together and 5 μmol.L-1 ranolazine.Radioimmunoasssay method was used to detect cell basic insulin secretion (BIS) and glucosestimulating insulin secretion (GSIS).TG in the cells was detected by enzyme assay and FAT/CD36 protein expression was determined by Western blot.Results In normal, high glucose and saturated fatty acid treatment and ranolazine treatment groups, the TG content was (2.31±0.05), (5.17±0.03) and(3.63±0.01) mmol.L-1, respectively.BIS in each group was (0.67±0.03), (0.32±0.07), (0.51±0.03) ng.mL-1; GSIS of each group was (1.68±0.17), (1.35±0.08), (1.47±0.11) ng.mL-1; FAT/CD36 relative expression ratio A1 in each group was (0.53±0.08), (1.78±0.05) and (1.07±0.06).As compared with the normal control, high glucose and saturated fatty acid could decrease the GSIS and increase the TG content and significantly increase the FAT/CD36 protein expression (P<0.05).But after co-treatment of high glucose, saturated fatty acid and ranolazine for 24 h, the GSIS was increased significantly, at the same time the intra-cellular TG content and the FAT/CD36 protein expression was decreased. Conclusion Ranolazine can protect the secretion function of NIT-1 cells impaired by high glucose and saturated fatty acid, and decrease TG content in the cells probably through the change of FAT/CD36 protein expression.

    Objective To study the effects of Biyun spray on allergic rhinitis (AR) induced by 2,4-tolunen diisocyanate (TDI) in guinea pigs. Methods TDI was selected as sensibiligen and administered intranasally to develop the AR model.The experimental guinea pigs were randomly divided into 4 groups: control group, model control group, Biyun spray group, positive control (ketotifen fumarate) group.Corresponding volume of 0.9% sodium chloride solution was administered to the control and model control groups.In the Biyun spray group, Biyun spray was administered at a dose of 96 mg.kg-1, which was twice the clinical equivalent dose.Ketotifen fumarate was administered to positive control group at a dose of 0.07 mg.kg-1.Symptoms were observed and the nasal symptom score of AR was determined.The pathological changes of nasal mucosa tissue were observed under a microscope.Eosinophilic granulocytes (EOS) and mast cells (MC) were counted.Results Among the control, model control, Biyun spray, and positive control groups, the nasal symptom scores of AR were decreased from (1.17±0.63), (7.05±1.24), (7.56±1.05), (7.69±0.96) to (1.11±0.54), (7.33±0.67), (4.92±0.96), (5.14±0.77), respectively after the treatments.Biyun spray significantly decreased the nasal symptom score of AR (P<0.01).Pathological changes of nasal mucosa tissue were improved obviously.Among the control, model control, Biyun spray, and positive control groups, the EOS number was (1.68±0.85), (9.13±1.68), (4.51±2.25) and (3.59±1.77), respectively; the MC number was (2.23±1.26), (21.03±5.03), (14.05±6.88) and (13.14±5.41), respectively; the degranulation MC number was (1.32±1.03), (13.64±5.47), (8.49±2.37) and (6.06±2.55), respectively.Biyun spray significantly improved the pathological changes of nasal mucosa with EOSs and MCs being decreased significantly (P<0.01).Conclusion Biyun spray is effective in treating allergic rhinitis in guinea pigs.

    Objective To investigate the relationship between imatinib withdrawal and recurrence in a patient with metastatic gastrointestinal stromal tumor (GIST) after surgery, and to figure out whether this kind of patients were still sensitive to imatinib after the second surgery. Methods The clinical characteristics before and after the recurrence were compared.The cell bio-markers of GIST in the patient before and after the recurrence were analyzed by immunohistochemistry.Meanwhile, KIT/PDGFR gene mutations was detected through DNA sequencing.Results The patient received a standard imatinib treatment for three years after surgery, and then he terminated this medical therapy by himself.About one year later, he suffered from an abdominal mass which was confirmed to be GIST recurrence by pathological examination.A KIT deletion mutation in exon 11 was detected: c.1667_1672delAGTGGA, which suggested that the patient was still sensitive to imatinib.Through comparing the pathological results , it showed that DOG1 was more sensitive than CD34.Conclusion Patients with GIST can benefit from continuous imatinib treatment by delaying the recurrence and prolonging the progressionfree survival time, and DOG1 is more sensitive than CD34 in diagnosis of GIST.

    Objective To explore the influence of curcumin on human Ewing’s sarcoma cell lines RD-ES in vitro. Methods The RD-ES cells were cultured in vitro and treated with different concentrations of curcumin solution for 24 to 72 h.MTT assay was performed to evaluate the cytotoxin effect of curcumin on the cells.TUNEL was used to detect apoptosis of the RD-ES cells.Bcl-2 expression was detected by immunofluorescence.Results Cell growth inhibitory rates were (11.8±2.0)%, (56.4±5.0)%, (67.6±6.0)% after treating with different concentrations of curcumins (1, 10 and 100 mg.mL-1, respectively) for 24 h, and the 50% inhibiting concentration was 10.74 mg.mL-1.After treatment of curcumins (1, 10 and 100 mg.mL-1, respectively) for 48 h, the cell apoptotic rate was increased with the increasing concentration of curcumins.The cell apoptotic rate was increased and Bcl-2 expression was decreased after treatment with curcumin.Conclusion Curcumin can induce RD-ES cell apoptosis, which is related with Bcl-2.

    Objective To study the antitussive mechanism of dextromethorphan. Methods By using reversedphase high performance liquid chromatography (RP-HPLC) with fluorescent light detector, we detected the effect of dextromethorphan on the contents of five kinds of monamine neurotransmitters in mouse brain with codeine phosphate serving as a positive drug.Results The results indicated that the antitussive mechanism of dextromethorphan was concerned with monanine neurotransmitter 5-HT.In the dextromethorphan group, 5-HT was 43.50% higher than in the blank group (P<0.01), and 5-HIAA was 35.00% higher than in the blank group (P<0.05).Dextromethorphan could increase the content of 5-HT in mouse brain to induce antitussive effects, while the antitussice effects of codeine phosphate had no relationship with serotonergic mechanisms.Conclusion The antitussive mechanism of dextromethorphan is related with the release of monanine neurotransmitter 5-HT in mouse brain.

    Objective To assess the clinical outcomes of periimplantitis treated with local administration of minocycline hydrochloride. Methods Twelve patients with one implant each and being diagnosed as periimplantitis were locally treated with minocycline hydrochloride after debridement and rinsing with 0.2% chlorhexidine solution.Plaque index, sulcus bleeding index, and probing depth were recorded at baseline and 4 weeks after the treatment, respectively.Results Plaque index, sulcus bleeding index, and probing depth were (2.42±0.79), (3.08±0.67), and (5.44±0.64) mm at baseline, and were (0.91±0.67), (1.33±0.88), and (3.98±0.78) mm at the forth week after the treatment, respectively.All differences were statistically significant (P<0.05).Conclusion The local administration of minocycline hydrochloride can be applied to treat periimplantitis effectively.

    Objective To study the best resins and the optimum separation conditions for obtaining highpurity total saponins from Xiezhuochubi decotion. Methods The best macroporous resin was selected by using static adsorption and desorption of total saponins, and the factors affecting the separation of total saponins were investigated.Results The macroporous resin D-101 not only presented the highest absorption capacity for saponins from the Xiezhuochubi decotion among the 6 resins studied, but also had a good desorption capacity.The optimum absorption conditions were as follows: the loading volume was 1.5 BV, absorption rate was 1.0 BV.h-1 and 30% ethanol could elute most of the saponins with the volume of 5 BV.The purity rate of saponins from the Xiezhuochubi decotion was up to 80% after the isolation and purification by macroporous resin D-101.Conclusion This method is simple and feasible and can meet the need of the largescale production.

    Objective To establish a finger print and quantitative determination of astilbin (A) and engelitin (E) simultaneously for Smilax china L.of different batches. Methods The samples were separated on a DIKMA Diamonsil C18 column with gradient elution of methanol and 0.1% phosphoric acid at a flow rate of 1.0 mL.min-1 and detected wavelength at 303 nm.Results The linear ranges of A and E were 0.08–20.52 μg (r=0.997) and 0.08–20.28 μg (r=0.998), respectively.The HPLC finger print of Smilax china L.of ten different places was established.Conclusion The method can be used for quality control of Smilax china L.

    Objective To study the conditions for isolating and purifying flavonoids in Tamarix chinensis Lour by D-101 type of macroporous adsorption resin. Methods The total flavonoids in Tamarix chinensis Lour was taken as a reference index.The sample volume, adsorption time, eluting velocity, ethanol concentration and pH value of the sample solution were investigated.Results The optimum conditions for isolating and purifying flavonoids in Tamarix chinensis Lour by D-101 type of macroporous adsorption resin were as follows: pH = 6-7, 15 mL sample of the original liquid, adsorption time of 30 min, 70% ethanol as an eluting reagent, and an eluting velocity of 3 BV.h-1.Conclusion D-101 macroporous adsorptive resin can effectively isolate and purify total flavones from Tamarix chinensis Lour.

    Objective To establish a method to determine content of ligustilide and cinnamylaldehyde in compound microemulsion of Radix Angelicae Sinensis (Danggui) (CMR). Methods High performance liquid chromatograph was used for detection.The column of Inertsil ODS3 (4.6 mm×150 mm, 5 μm) was used and the detection wavelength was 286 nm.The mobile phase was Acetonitrile-water (55:45) at a flow rate of 1.0 mL.min-1 and the temperature of column was set at 30 ℃.Results The calibration curve of Cinnamaldehyde was linear in the range of 0.484 5-38.75 μg.mL-1.The regression equation was Y=27.06X-1.586 (r=0.999 8).The recovery rate was 99.60%.The intra-day and inter-day precision was good with the RSD of 1.251% or below.The calibration curve of ligustilide was liner in the range of 0.531 0-106.1 μg.mL-1.The regression equation was Y=13.93X+1.344 (r=0.999 8).The recovery rate was 98.94%.The intraday and interday precision was good with the RSD of 1.25% or below.Conclusion This method is convenient, rapid and accurate.It can be used for quality control of CRM.

    Objective To establish a method for identification of Caulis Polygoni Multiflori in Jiannao mixture and determination of emodin. Methods Emodin, as an effective constituent of Caulis Polygoni Multiflori in preparation, was identified by thinlayer chromatography (TLC), and the content was determined by high performance liquid chromatography (HPLC).Results The characteristics of the active ingredient of Caulis Polygoni Multiflori could be identified by TLC.The linear range of emodin was within 0.053 7-215.000 0 μg.mL-1(R2≥0.999 7).The RSD of intraday and interday precisions were not more than 1.53% (n=6).The average recovery rates of the investigated compound at low, medium and high concentration levels were in the range of 98.83%-99.53% for all with RSD no more than 1.33% (n=6).Conclusion The developed method is simple, rapid, sensitive and accurate with good repeatability.It can be used for quality control of Jiannao mixture.

    Objective To establish a quality standard of zicao jiedu ointment. Methods Berberine hydrochloride and Radix Arnebiae were identified by thinlayer chromatography (TLC).The content of L-shikonin was determined by high performance liquid chromatogram (HPLC).The column was Hypersil BDS C18 column (4.6 mm×250 mm, 5 μm).The mobile phase consisted of methanol0.2% phosphoric acid aqueous solution (25:75).The detective wavelength was 516 nm, flow rate was 1.0 mL.min-1, and the column temperature was 30 ℃.Results TLC was a good method to separate Berberine hydrochloride and Radix Arnebiae.The calibration curve of L-shikonin was linear in the range of 4.47-143.10 μg.mL-1 with good linearity (r=0.999 9).The average recovery was 97.5% (RSD=1.42%).Conclusion The method is simple, accurate and sensitive.It can be used for quality control of zicao jiedu ointment.

    Objective To determinate the contents of hypericin and hyperoside in Hypericum sampsonii Hance by reversedphase highperformance liquid chromatography (RP-HPLC). Methods HPLC was performed on DiamonsilTMC18 (200 mm×4.6 mm, 5 μm) with methanol-ethylactate-1.0% sodium dihydrogen phosphate (340:15:5) and methanol-0.025 mol.L-1 phosphatic acid (50:50) as mobile phase.Detection wavelengths were 588 nm and 360 nm respectively.Results The linear ranges of hypericin and hyperoside were 0.02-0.18 μg and 0.05-0.80 μg, respectively.The recovery rates of them were respectively 98.08% (n=6)and 97.58% (n=6), RSDs were 1.05% and 0.89%, respectively.Conclusion The method was accurate and highly reproducible, and can be used for the quality control of Hypericum sampsonii Hance.

    Objective To prepare compound amoxicillin capsules and investigate the method of quality control. Methods Wet granulation was used to prepare the compound amoxicillin capsules and high performance liquid chromatography (HPLC) was used to determine the content and dissolution.Results The calibration curves were linear in the ranges of 80-320 μg.mL-1 for amoxicillin and 4.8-19.2 μg.mL-1 for ambroxol.Their average recovery rates were 96.72% and 104.17%; RSDs were 1.84% and 0.57%, respectively.The content and dissolution of samples of three batches were all above 90% and 80%.Conclusion The content and dissolution of the compound amoxicillin capsules are both consistent with the Standard of Pharmacopoeia of China.This HPLC method is accurate, simple and effective for separation, and can be used for quality control of the compound capsules.

    Objective To extract total alkaloids of Sinomenium acutum by Soxhlet extractor and analyze them by gas chromatography-mass spectrometry (GC-MS). Methods The Sinomenium acutum was extracted by 90% alcohol, acidified by hydrochloric acid, alkalinized by ammonia water, and then extracted by chloroform.Fifteen peaks were separated and identified by GC-MS.Results The relative content of sinomenine, ambelline and stepharine was up to 80.18%, 3.44% and 3.05%, respectively.The compounds were quantitatively determined by normalization method, and the identified compounds represented 95.1% of total GC peak area.Conclusion The results provide important foundation to further study and exploit Sinomenium acutum.

    Objective To establish gas chromatography (GC) method for separating and determining both borneol and menthol in tanxiang tongmai capsule. Methods Naphthalene served as an internal standard, acetic ether as a solvent, 10% PEG-20M as separation, nitrogen as the carrier gas.Column temperature was set at 130 ℃, and injector and detector temperature was 180 ℃.Sample size was 1 μL by using flame ionization detector.Results The linear ranges of borneol and menthol were 0.26-2.55 mg.mL-1 (r=0.999 9) and 0.26–2.62 mg.mL-1 (r=0.999 8), respectively.Recovery rates of borneol and menthol were 97.5% (RSD=1.77%) and 96.81% (RSD=1.28%), respectively.Conclusion This method is convenient, rapid, accurate and reproducible, and can be used for quality control of tanxiang tongmai capsule.

    Objective To study the stability and compatibility of lornoxicam and morphine hydrochloride in 0.9% sodium chloride injection. Methods In 0.9% sodium chloride injection, the content change of lornoxicam and morphine hydrochloride was determined by reversed phased high performance liquid chromatogram under room temperature (20±1) ℃ within 72 h, and the appearance of the solution was observed and its pH value was determined.Results No significant differences were found in the content, color and pH value for the mixed solution at different time points within 72 h after mixing at room temperature.Conclusion The mixture of lornoxicam and morphine hydrochloride in 0.9% sodium chloride injection at ambient temperature was stable within 72 h.

    Clinical pharmacists participated in a treatment of the patient with lung cancer and malignant pleural effusion (MPE).The antiinfective therapy, chemotherapy and management of MPE were analyzed retrospectively.The approaches for clinical pharmacists participating in pharmaceutical care on a patient with lung cancer and MPE were explored.Through pharmaceutical care for the patient, the symptoms of infection and MPE were alleviated and severe adverse drug reactions were avoided.Individual pharmaceutical care on patients with lung cancer and MPE can improve therapeutic efficacy and life quality of the patient.
