    Objective To study the effects of medicated serum with total saponins from Rhizoma Dioscreae Nipponicae (RDN) on VEGF mRNA expression and AP-1 activity in rat synovial cell strain RSC-364 induced by IL-17 and TNF-α.To investigate the mechanism about total saponin from RDN inhibition of angiogenesis. Methods Medicated serum of total saponins from RDN and tripterygium (positive control) were prepared.Rat synovial cells RSC-364 were divided into four groups: the blank control,IL-17+TNF-α model,tripterygium medicated serum,and total saponins medicated serum groups.After one hour of incubation,all groups except for the blank control were incubated with both IL-17(10 μg·L-1) and TNF-α(10 μg·L-1) for 24 hours.VEGF mRNA expression in RSC-364 was detected by PrimeScriptTM real-time quantitative PCR (RT-PCR) detection kit,and the AP-1 DNA-binding activity was detected by electrophoretic mobility shift assay (EMSA). Results Compared with the control blank group,both of the VEGF mRNA expression and AP-1 activity in rat synovial cell strain RSC-364 induced by IL-17 and TNF-α increased remarkably (P<0.05,P<0.01).The VEGF mRNA expression and AP-1 activity in tripterygium medicated serum group and total saponins medicated serum group were remarkably lower than those of the model control group (P<0.05).There was no significant difference between the two medicated serum groups. Conclusion Serum medicated with total saponins from RDN can remarkably decrease VEGF mRNA expression and AP-1 activity,indicating that the total saponins from RDN could influence VEGF secretion by inhibiting the AP-1 signal transduction pathway,VEGF is the key factor of angiogenesis,thereby to restrain angiogenesis.

    Objective To study the therapeutic effects and related mechanism of Tongmai Yizhi granules on vascular dementia in rats. Methods Bilateral carotid artery ligation was used to establish rat model of vascular dementia (VD).The rats were randomly divided into 6 groups: sham operated,the model control,piracetam (0.375 g·kg-1·d-1) as positive control,low dose (2.5 g·kg-1·d-1),middle dose(5.0 g·kg-1·d-1),and high dose (10.0 g·kg-1·d-1) of Tongmai Yizhi granules.Each group was intragastrically administered with 10 mL·kg-1 of corresponding medications for 31 days after the VD model was established.The sham group was given with 0.9% NaCl solution.Y-electric maze was used to test the learning and memory function of rats at the second and fourth weeks.On the day 32,hippocampal tissues were collected for pathological analysis by microscope.The activities of SOD and content of MDA in serum and tissue homogenate were tested. Results Compared with model control group,each dose of Tongmai Yizhi granules obviously shortened the incubation period of VD rats in electric maze test,and accuracy of learning and memory was improved (P<0.05 or P<0.01).The activity of SOD was decreased,and the content of MDA was increased after treatment with Tongmai Yizhi granules (P < 0.01).Hippocampal neuronal cells were disarranged,and the number of cone cells was decreased significantly in the model control group.In contrast,the necrosis and degeneration of hippocampal neurons were alleviated in all Tongmai Yizhi treatment groups. Conclusion Tongmai Yizhi granule can apparently improve learning and memory function of VD rats.It can also improve the activity of SOD and reduce the content of MDA in serum and hippocampus,effectively remove free radicals,and alleviate the injury of hippocampal pyramidal cells.

    Objective To investigate the reversal effect and molecular mechanisms of NF-κB inhibitor parthenolide (PTL) on insulin resistance (IR). Methods HepG2 cells were treated with insulin at different concentrations and time points, the glucose consumption of HepG2 cells was measured via glucose oxidase method to determine the best concentrations and time for establishing insulin-induced insulin resistance on HepG2 cells.After modeling, different concentrations of PTL were added in cells for 24 h for determining cell activity and glucose-consumption.Q-PCR was used to detect the expression of NF-κB and IκBα mRNA in cells, and western blot was used to detect the expression of protein IκBα. Results The best reaction time and concentration for insulin inducing resistance of HepG2 cells were 24 h and at 10 μg·mL-1.The optimum acting dose of PTL was 20 μmol·L-1.NF-κB activity was significantly reduced (P<0.05), IκBα degradation was significantly inhibited (P<0.05) compared to HepG2 cells with insulin resistance upon intervention on insulin resistance HepG2 cells by PTL. Conclusion PTL can inhibit IκBα degradation and disassociation of it from NF-κB, which in turn improves insulin resistance, providing theoretical basis for preventing and treating diabetes with PTL.

    Objective To investigate the anti-inflammatory and analgesic effects of extracts from Garden Balsam seeds in order to provide theoretical basis for rational clinical application. Methods The mouse ear swelling induced by xylene, the body torsion induced by acetic acid or hot plate method, the rat foot swelling induced by egg white and granuloma model were set up to study anti-inflammatory and analgesic effects of extracts from Garden Balsam seeds. Results The water extracts at the middle and high doses reduced the body torsion counts of mice (P<0.05), alleviated the rat toe swelling, but did not affect mice ear swelling.The extracts at the high dose prolonged pain threshold at 60 min (P<0.01) and 90 min (P<0.05); the alcohol extracts at the middle and high doses significantly lowered body twisting counts (P<0.01), attenuated the ear swelling of mice (P<0.05), remarkably prolonged the pain threshold (P<0.01)at 30 min, weakened the rat foot swelling, and elevated 60 min pain threshold at the high doses (P<0.01), but the extracts exerted no effects on the rats granuloma. Conclusion The extracts from Garden Balsam seeds have anti-inflammatory and analgesic effects.

    Objective To investigate the effect of acteoside on injury PC12 cells induced by glutamate. Methods PC12 cells were assigned into normal control group, model control group, positive drug group and acteoside(AS) treated group.Every group was treated by 1.5 mmol·L-1 glutamate for 24 hours except for the control group, and the injury was antagonized by 10 μmol·L-1 Vit E and acteoside at different concentration(15.625, 31.25, 62.5, 125 and 250 μmol·L-1).Cell morphology was observed by inverted microscope, cell survival was determined with MTT, LDH activity was measured by enzyme label kit, the MDA content and SOD activity were measured by TBA kit and WST kit, and the ROS was measured by Elisa kit. Results Compared with the model control group, all doses of acteoside could significantly improve the PC12 cell morphology and survival(P<0.05), inhibit LDH activity and production of MDA and ROS (P<0.05), increase the activity of SOD (P<0.05), except for the lowest dose group. Conclusion Acteoside has protective effects on PC12 cells injured by glutamate.

    Objective To study the inhibitory effect and mechanism of berberine on the malignant potential of liver cirrhosis tissue due to schistosoma. Methods Eighty rats infected with schistosoma japonicum were equally divided into A, B, C and D groups, twenty rats in each.Group A was taken as model control without any treatment.Group B was treated with praziquantel at 500 mg·kg-1·d-1 for 2 days.Group C was treated with berberine at 150 mg·kg-1·d-1 for 6 weeks.Group D was treated with berberine at 150 mg·kg-1·d-1 for 6 weeks followed by praziquantel treatment at 500 mg·kg-1·d-1 for 2 days.Ten rats were taken as normal control group (group E).At 12th week, all rats were sacrificed and parts of liver tissues were preserved.The contents of IFN-γ,typeⅠ collagen, type Ⅲ collagen, telomerase reverse transcriptase (TERT )and the activity of telomerase in liver tissue before and after treatments were detected. Results Compared to the group A, the therapy of praziquantel or berberine singly reduced the contents of typeⅠand type Ⅲ collagens,TERT and the activity of telomerase and greatly raised the content of IFN-γ(P<0.05 ).Compared to the group B, there was no difference on the contents of IFN-γ,typeⅠ collagen,type Ⅲ collagen,TERT and the activity of telomerase in liver tissue between two groups(P>0.05).Compared to the group C or group B, the contents of typeⅠ collagen,type Ⅲ collagen,TERT and the activity of telomerase in liver tissue were obviously reduced and the content of IFN-γ was conspicuously elevated (P<0.05 ) in the group D. Conclusion Berberine can inhibit the malignancy by inhibiting the expression of TERT and the activity of telomerase via regulating immune response and promoting the expression of IFN- γ in liver tissue.Berberine may be used in symergism with the insecticidal treatment for advanced schistosomal hepatic cirrhosis.

    Objective To investigate the effect of rosiglitazone on the epithelial-mesenchymal transition process of renal tubular epithelial cell induced by transforming growth factor-β(TGF-β). Methods The human renal tubular epithelial cell line HK-2 was cultured in vitro and treated with TGF-β at the presence or absence of rosiglitazone, and the morphological changes of HK-2 cells were observed.Then PPARγ,SMAD family number 2/3,E-cadherin and Vimentin were detected by western blot, and the mRNA expression of PPARγ, E-cadherin, Vimentin, zinc finger transcription factor Snail and Slug were measured by realtime quantitative PCR.Finally,dual luciferase reporter assay was performed to detect the effect of TGF-β and rosiglitazone on the transcription activity of E-cadherin promoter. Results TGF-β could induce the EMT process of HK-2, including morphological changes such as longer pseudopod and broader cell space.TGF-β also activated the SMAD2/3 signaling pathway, with expression changes of E-cadherin and Vimentin.Rosiglitazone reversed the morphological changes of HK-2 cells induced by TGF-β, with shorter pseudopod and narrower cell space.Rosiglitazone also inhibited the mRNA and protein expressions of Vimentin, and restored the mRNA and protein expression of E-cadherin by up-regulating PPARγ.The activity of E-cadherin promoter was enhanced under the treatment of rosiglitazone. Conclusion Rosiglitazone can antagonize the EMT process of renal tubular epithelial cell induced by TGF-β, through the activation of PPARγ and stimulating transcription and protein expression of E-cadherin.

    Objective To investigate the scavenging effect of Baoyuan anticancer oral solution on superoxide anion(O2) and 1,1-diphenyl-2-picrylhydrazyl hydrate(DPPH) free radicals. Methods O2 and DPPH were examined with the ESR (electron spin resonance) spin trapping method. Results The scavenging effect of Baoyuan anticancer oral solution diluted to 50 fold, on O2 and DPPH free radicals was 81.32% and 98.47%, respectively. Conclusion Baoyuan anticancer oral solution has a dose-related effect on free radical elimination.

    Objective To study the expression of insulin like growth factor binding proteins 3 (IGFBP-3) during inhibition of resveratrol (Res) on cell proliferation. Methods The inhibitory effect of Res on BGC-823 cells was determined by MTT method; Real-time qRT-PCR and western blot were applied to detect the expression of IGFBP-3 in Res-treated BGC-823 cells.In addition, cytometry was used to determine the proliferation and apoptosis of Res-treated BGC-823 after knockdown of IGFBP-3 by siRNA. Results Upon Res (20,40, 80 and 160 μmol·L-1) treatment,the viability of BGC-823 cells was (82.35±10.65)%,(74.30±12.36)%,(62.80±14.66)% and (50.75±11.14)%, respectively.The mRNA and protein expression of IGFBP-3 elevated as high as 2.96-fold compared to the control group (P<0.05).The cell viability of BGC-823 cells with IGFBP-3 knockdown was significantly higher than that of the wild type (P<0.05) only at high Res concentration (160 μmol·L-1).Meanwhile,IGFBP-3 knockdown led to a significant decrease on cell apoptotic rate by Res (160 μmol·L-1) [(20.13±9.12)% vs (35.48±11.12)%,P<0.05)]. Conclusion Res can inhibit BGC-823 cell proliferation and promote cell apoptosis, the underlying mechanism of which may be related to the overexpression of IGFBP-3 in BGC-823 cells.

    Objective To evaluate the effects of pregabalin combined with hydrochloric oxycodone on patients with malignant neuropathic pain (MNP). Methods A total of 66 patients with MNP was divided into control group or treatment group randomly.The patients in control group received only hydrochloric oxycodone, and treatment group were treated with the combination of pregabalin and hydrochloric oxycodone.Numeric rating scale (NRS) score was used to evaluate the analgesic effects.Medical outcomes study sleep scale (MOS-SS,Chinese version) was used to evaluate the improvement of sleep disorder.The changes of depression or anxiety were investigated by 17-item Hamilton Depression Rating Scale (HAMD-17) or Hamilton Anxiety Scale (HAMA), respectively.Side effects were accessed by Acute and Subacute Toxicity Grading Criteria of Anticancer Drugs (WHO). Results The pain control rate of treatment group was 87.1%, which was superior to that of control group (58.6%) (P<0.05).The improvement of sleep interference, and the quality and quantity of sleep in treatment group were also superior to that in control group (P<0.05).After the treatment, depression and anxiety was attenuated in both groups, and the improvement degree in treatment group was higher than that in control group (P<0.05).No obvious side effects were found in either groups. Conclusion The combination therapy of pregabalin and hydrochloric oxycodone is the better way to treat MNP.

    Objective To assess the clinical efficiency and side effects of topical mometasone furoate aqueous nasal spray for patients with naphazoline-induced rebound rhinitis. Methods A prospective, non-randomized and self-controlled study was applied.A total of 22 patients with naphazoline-induced rhinitis received two spray-puffs of 50 μg mometason furoate twice daily to each nostril in the morning for one week, followed by two spray-puffs of 50 μg mometason furoate once daily to each nostril every morning for another three weeks after withdrawn of naphazoline therapy.Nasal congestion was evaluated by visual analogue scale (VAS) recorded before and after treatment.Side effects like nose bleeding and perforation of the nasal septum was detected with rhinoscope. Results The statistical difference of VAS before and after treatment was significant.No adverse reactions as nose bleeding and perforation of the nasal septum were observed by the end of treatment. Conclusion It is safe and effective to use mometasone furoate aqueous nasal spray for treating rebound rhinitis induced by misuse of naphazoline nasal drop.

    Objective To evaluate the effect of neoadjuvant chemotherapy by different arteriovenous ways on ovarian function of patients with locally advanced cervical cancer after surgery. Methods We retrospectively studied 38 cases of young women (surgical age <45 years) who were diagnosed with locally advanced squamous cell carcinoma of the cervix and undergone radical hysterectomy with ovaries.According to the way of adjuvant chemotherapy, these cases were divided into 20 cases of group A (arterial embolization chemotherapy), 18 cases of group B (intravenous chemotherapy).Each patient underwent a course of chemotherapy.Then we retrospectively analyzed and compared serum estradiol (E2), follicle stimulating hormone (FSH), luteinizing hormone (LH) in two groups before and after surgery to assess ovarian function.We compared E2,FSH and LH levels 2-3 weeks after chemotherapy (before surgery) and 1 month after surgery,and 6 months after surgery in two groups. Results The E2 levels in group A 1 month before and after surgery significantly decreased, while FSH increased compared with those before chemotherapy(P<0.05).LH levels in two groups at different time points and 6 months after surgery showed no significant difference (P>0.05) compared with those before chemotherapy.the E2, FSH and LH levels in group B at different time points presented no significant difference (P>0.05).Only E2 level in group A was significantly lower than that in group B before surgery. Conclusion For patients with locally advanced cervical cancer, the effect of arterial embolization chemotherapy before surgical intervention on ovarian function is transient and reversible, while intravenous chemotherapy before surgery exerts few effects on ovarian function.Retaining ovaries in radical hysterectomy is a safe and effective method to protect the endocrine function for young patients with locally advanced cervical cancer.

    Objective To establish the quality control method for Antenoron filiforme. Methods Antenoron filiforme was identified by TLC,and the content of gallic acid was determined by HPLC.The chromatographic conditions were as follows: Aichrom Reliasil C18 column (4.6 mm×250 mm,5 μm) was used with mobile phase consisted of acetonitrle-0.1% phosphoric acid(4:96),the flow rate was 1.0 mL·min-1,and the detection wavelength was 270 nm. Results TLC identification for Antenoron filiforme was highly specific.Gallic acid content had a good linearity in the range of 0.08-0.56μg (r=1.000 0).The average recovery was 101.12% and RSD was 2.20% (n=6). Conclusion The method is simple, feasible,and reproducible,and can be used for the quality control of Antenoron filiforme.

    Objective To compare transdermal penetration of triamcinolone acetonide liposparticles (TAA-LPPs) and TAA-Ethosomes in vitro. Methods The TAA-LPPs and TAA-Ethosomes were produced and the morphology was observed by transmission electron microscope,particle size was detected by laser particle analyzer.The percutaneous permeability in vitro was tested by modified Franz diffusion pools.The amount of penetrated triamcinolone acetonide and the retention in the skin were determined by HPLC. Results The shape of TAA-LPPs and TAA-Ethosomes was almost spherical with mean diameter of (99.9±1.3) and (105±1.4) nm, respectively.The cumulative transdermal penetration of TAA-LPPs, TAA-Ethosomess and TAA suspension was (53.59±4.40),(87.03±4.87),and (30.54±8.61) μg·(cm2)-1, respectively .The drug retention in the skin after 32 h was (1.02±0.13), (0.62±0.08), (0.55±0.17) μg·(cm2)-1, respectively. Conclusion TAA-LPPs is better for transdermal administration of triamcinolone acetonide by reducing systemic absorption of the drug.

    Objective To optimize preparation of freeze-dried 16-dehydropregnenolone (16-DHP) liposome and evaluate the quality. Methods The freeze-dried liposome was prepared by rotary-evaporated film-ultrasonication with lyophilization.Single factor experiments and orthogonal design were used to optimize the prescription and preparation,and the quality was evaluated. Results The optimum preparation technics were as follows: the concentration of PC-98T was 6.5% (W/V), the quantity ratio of PC-98T to cholesteryl sodium sulfate salt was 4:1,hydration temperature was 60 ℃, ultrasonic time was 2 min,and the cryoprotectant was 7.5% glucose.The average particle size, Zeta potential and encapsulation efficiency were (121.3±48.7) nm,-41.9 mV and (98.7±0.1)%, respectively. Conclusion The formulation of freeze-dried 16-DHP liposome is reasonable,the technology is practicable and stable.

    Objective To establish the quality control of Yinhuang tablet (YHT) by using sever ingredients determination and tri-wavelength fingerprint analysis technologies. Methods The chromatographic fingerprints were developed by using Agilent Poroshell 120 SB C18 (4.6 mm×150 mm, 2.7 μm) as the separation column and 0.2% H3PO4-methanol as the mobile phase.The flow rate was 0.5 mL·min-1, the UV absorbance was monitored at 214, 278 and 326 nm by injecting 5 μL of the sample solution and the column temperature was maintained at (35.00±0.15) ℃.The chromatographic fingerprints were characteristically evaluated for quality ranking of 13 batches of YHT by the 4.0 software of the digitized evaluation system of traditional chinese medicine fingerprint with the super information characteristics.Seven marker compounds including chlorogenic acid, caffeic acid, baicalin, wogonoside, baicalein, wogonin and scutellarin were simultaneously determined to compare the variation among five different manufacturers. Results The tri-wavelength fingerprint high performance liquid chromatography method was established.A total of 28(214 nm),29(278 nm)and 26(326 nm)common peaks were identified at respective wavelengths, using baicalin (BCL) as the referential peak.The software was applied to evaluate the YHT-HPLC fingerprints and identify the quality of 13 batches of YHT by systematically quantified fingerprint method (SQFM).The results showed that the content from the different manufacturers varied greatly. Conclusion The quality of YHT varies between different makers, and is necessary to establish the criterion for quality control.This method can provide a new reference for the quality control of YHT.

    Objective To establish a method for determining cinnamic aldehyde content in Xianggui Huazhuo capsules by gas chromatography-mass spectrometry (GC-MS). Methods The content of cinnamic aldehyde was determined by GC-MS.Separation was performed on a capillary column (30 m×0.25 mm, 0.25 μm) with HP-5 as the stationary phase.A programmed temperature was employed.The flow rate was 1 mL·min-1 with He as carrier gas, and split ratio was 50:1.The injection volume was 1.0 μL. Results The cinnamic aldehyde was well isolated from the other ingredients.A good linear relationships of cinnamic aldehyde in range of 0.02-4.00 mg·mL-1 was observed.The correlation coefficient was 0.999 4.The average recovery of cinnamic aldehyde was 96.2%, and RSD was less than 2.11%. Conclusion The method is simple, accurate and suitable for determination of cinnamic aldehyde content.

    Objective To prepare vinorelbine bitartrate long-circulation liposomes by pH gradient loading methods and make characterization. Methods The impact of hydration temperature and extrusion times on the blank liposome particle size was investigated;and the incubation temperature and in duration on size and encapsulation percentage of drug loading liposome particle was tested.The vinorelbine bitartrate long-circulation liposome was characterized for particle size,polydispersion index,Zeta potential,morphology,and was studied for long term stability. Results The particle size,Zeta potential,polydispersion index of long-circulation liposomes were (96.4±27.2) nm,(0.162±0.042),(-26.7±3.5) mV,respectively.The liposomes were small,unilamellar and spherical with smooth surface under transmission electron microscopy.Long term stability studies showed that the liposomes were stable for up to 3 months after storage at 5 ℃. Conclusion The preparation technology for the vinorelbine bitartrate long-circulation liposome by pH gradient loading methods is feasible.

    Objective To develop a HPLC method for determination of aesculin, aesculetin,baicalin and ellagic acid in Xielining tablets. Methods The hypersil C18 column was used with the flow rate of 1.1 mL·min-1.The mobile phase A consisted of methanol-acetonitrile(4:1),the mobile phase B consisted of 0.1% phosphoric acid solution; The detection wavelengths were λ1=334 nm(aesculin and aesculetin),λ2=280 nm(baicalin),and λ3=254 nm(ellagic acid). Results There was a good linear relationship between the peak area values and concentrations of aesculin,aesculetin,baicalin and ellagic acid.The quantitation range of aesculin,aesculetin,baicalin and ellagic acid was 0.058 6-1.172 0 μg(r=0.999 2), 0.015 4-0.308 0 μg(r=0.999 8),0.447 2-8.944 0 μg(r=0.999 6),and 0.072 6-1.452 0 μg(r=0.999 5), respectively.The average recovery was 97.24%(RSD=0.78%),97.76%(RSD=1.11%),98.43%(RSD=0.93%) and 96.89%(RSD=0.62%), respectively. Conclusion The method is convenient,accurate,sensitive,reproducible and may be used in the determination of aesculin, aesculetin,baicalin and ellagic acid in Xielining tablets.
