As a famous herb for treating blood stasis, Salvia miltiorrhiza Bunge (Danshen) has a long history of medical application. Various formulations of Danshen related drugs [derivatives from Danshen and traditional Chinese medicine (TCM) formula including Danshen] have been widely used in clinical practice. However, with the increase of applications and the expansion of indications, reports on their adverse drug reactions are dramatically increased. This article briefly introduced the incompatibility and use notes of Danshen in ancient TCM books. Furthermore, the adverse drug reactions and incompatibility in modern clinical practice were reviewed and some strategies to avoid and/or handle these adverse effects were proposed. We hope to provide reference for Danshen-based new drug research and development, and for clinical application of Danshen safely and effectively.

Because of limit knowledge for biological underpinning of depression, current antidepressant mainly focuses on attenuating the pathological changes and neuropsychiatric symptoms. Emerging evidence indicates the importance of adult hippocampal neurogenesis in regulating the adaptability to psychiatric or psychological stress. Additionally, increasing hippocampal neurogenesis is an effective approach for antidepressant therapy. Abnormalities of metabolic factors in contributing the development of neurological or psychiatric diseases have been widely documented. Investigating the function of metabolic factors could offer the new target for antidepressants exploration. In this review, we summarized the functions of different types of metabolic factors in regulating neurogenesis as well as the possibility of these factors as the antidepressant targets. Meanwhile, we introduced the previous work in our laboratory: the roles of metabolic factor APPL2 in regulating adult hippocampal neurogenesis and antidepressant behaviors. By targeting APPL2, baicalin could exert the neurogenic promotion in hippocampus and olfactory system and thereby attenuate the emotional and olfactory dysfunctions in depression model. This case offers the significance of metabolic factor as the effective targets in regulating neurogenesis and antidepressant. Moreover, by targeting these identified metabolic factors, many types of traditional Chinese medicine monomers may be applied as the effective antidepressants.

Objective To observe the effect of oleuropein on cognitive function of aged rats received splenectomy. Methods Pathogen-free healthy male aged SD rats were divided into 4 groups using a random number table: sham group, surgery group, oleuropein group ( low dose group),oleuropein group (high dose group). Morris water maze was performed to detect cognitive function in the rats. The levels of malondialdehyde (MDA) and superoxide dismutase (SOD) in the hippocampus were determined by ELISA. The p-ERK1/2,Nrf2 and nuclear Nrf2 (n-Nrf2) was detected by Western blotting. Results There was no significant difference in the latency, the number of platform-site crossovers, and the time spent in target quadrant of each group before surgery. Compared with the sham group, the latency of surgery group was significantly extend, meanwhile, the number of platform-site crossovers and the time spent in target quadrant were significantly reduced. In the oleuropein group, the latency was shortened (P<0.05),the number of punctures increased (P<0.01),and swimming time increased (P<0.01) comparing to surgery group. After surgery, the content of MDA in the hippocampus of surgery group was significantly increased, and the SOD content was significantly decreased. In the oleuropein group, the content of MDA decreased (P<0.01),and SOD increased (P<0.05, P<0.01) comparing to surgery group. Western blotting showed, the p-ERK1/2,the n-Nrf2 and total Nrf2 levels in the surgery group were significantly decreased when compared with the sham group. In the hippocampus of oleuropein groups, the expression of p-ERK1/2 (P<0.05, P<0.01),n-Nrf2 (P<0.01) and the total Nrf2 (P<0.01) significantly increased comparing to surgery group. Conclusion Oleuropein can significantly improve cognitive impairment in elderly rats after splenectomy, and its mechanism may be related to its inhibitory effect of Nrf2/ERK pathway.

Objective To observe oral toxicity of zilanine in rats and to evaluate its safety. Methods One hundred and sixty rats were randomly divided into 4 groups: control group, low, middle and high dose groups of zilanine. The rats were orally administrated with zilanine of 20,200 and 2000 mg·kg-1·d-1 for six months, respectively. The general condition, body weight, food consumption, water consumption, hematology, blood biochemistry test, organ coefficients and histopathological changes of all the rats were observed. Results There was no obvious effect on the general condition, food consumption, water consumption, organ coefficients in all the rats treated with zilanine. The middle dose zilanine caused an increase of neutrophils and a decrease of the number of lymphocytes. Rat body weight of high dose group was significantly lower than the control group. High dose of zilanine caused the increase of neutrophils, and the number of lymphocytes, the erythrocyte, hemoglobin, and eosinophil. The total protein (TP),creatinine (Cr),and creatine phosphokinase (CKP) decreased in rats. All of these parameters restored to baseline level after 4 weeks. No toxic pathological changes were observed in the high, middle and low dose groups, and no delayed toxic reactions were observed after 4 weeks of drug withdrawal. Conclusion The dose of 20 mg·kg-1·d-1 of zilanine shows non-toxic response at this study condition.

Methods Healthy adult male SD rats, were used in experiments. Thirty-six successfully prepared Langendorff isolated cardiac perfusion models were randomly divided into 6 groups (n=6 in each group) by random number table. After perfused with 37 ℃ K-H solution for 15 mins, the control group (group C) continued to perfusing with 37 ℃ K-H solution for 30 mins; the 35 ℃ hypothermia group (group H1) and the 32 ℃ hypothermia group (group H2) continued to perfusing with 35 ℃ or 32 ℃ K-H solution for 30 mins. the sevoflurane group (group S) continued to perfusing with 37 ℃ K-H solution containing 1.0 MAC sevoflurane saturation for 30 mins; the 35 ℃ hypothermia combined with sevoflurane group (group H1S) and the 32 ℃ hypothermia combined with sevoflurane group (group H2S) was continuously perfused with 1.0MAC sevoflurane saturated 35 ℃ K-H solution and 32 ℃ K-H solution for 30 mins. At the end of equilibrium perfusion (t0) and continuous perfusion 30min (t1),the monophasic action potential (MAP) was recorded and the myocardial MAP duration at 90% repolarization (MAPD90) was calculated. At t1, the ERP and the ratio of ERP to MAPD90 (ERP/MAPD90) were measured and calculated. Regular stimuli (S1) were followed by a single extra-stimulus (S2),the longest pacing cycle length (PCL) of ventricular fibrillation threshold (VFT) was recorded. And the occurrence of arrhythmia was recorded during the whole perfusion. Results MAPD90 was prolonged in group H2 and group H2S at t1 when compared with that of t0 (P<0.05). At t1,MAPD90, ERP and the logest PCL of VFT of group H2 were prolonged, and ERP/MAPD90 ratio of group H2 was decreased comparing to those of group C or group H1 (P<0.05). MAPD90,ERP and the longest PCL of VFT were prolonged at t1 in group H2S (P<0.05),but there was no significant difference in ERP/MAPD90 ratio (P>0.05). Compared with group S,MAPD90,ERP and the longest PCL of VFT were prolonged at t1 in group H2S(P<0.05),but there was no significant difference in ERP/MAPD90 ratio(P>0.05). Compared with the group H2,MAPD90 and ERP were shortened,ERP/MAPD90 ratio was increased, and the longest PCL of VFT was shortened in group H2S (P<0.05). The rats of ventricular arrhythmia were fewer in group H2S than in group H2 (P<0.05). Conclusion Sevoflurane can inhibit the decrease of electrophysiological stability of ventricular myocardium induced by hypothermia and reduce the risk of ventricular arrhythmia.

Objective This study aimed to evaluate the effects of 3,3'-diindolylmethane (DIM) on LPS-induced inflammatory injury in neonatal rat cardiomyocytes and explore its molecular mechanisms. Methods The neonatal rat cardiomyocytes were isolated and cultured, which was divided into 4 groups: control group,LPS (10 mg·L-1) group,DIM (20 μmol·L-1) group and LPS+DIM group. Cell viability was measured by CCK8,and the morphology of cultured neonatal rat cardiomyocytes was observed with immunofluorescence staining. The mRNA expression levels of pro-inflammatory mediators were measured with real-time PCR and the protein expression levels were examined with Western blotting analysis. Results The mRNA levels of early inflammatory factors in the LPS + DIM group, including interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-α) and late pro-inflammatory cytokine high mobility group box 1(HMGB1),were significantly lower than those of the LPS group (P<0.05). Moreover,in the LPS + DIM group, the elevated levels of proteins such as Toll like receptor-4 (TLR-4) and phosphorylated extracellular signal-regulated kinases 1/2 (ERK1/2),phosphorylated P38 and phosphorylated c-Jun NH2-terminal kinase (JNK) induced by LPS were significantly lower than those of the LPS group. Conclusion DIM had a protective effect on LPS-induced inflammatory injury in neonatal rat cardiomyocytes, and modulated the excessive production of pro-inflammatory cytokines IL-6,TNF-α and HMGB1 via suppressing the TLR-4/MAPKs signaling pathways.

Objective To evaluate the potential impact of hawthorn fine powder on the cardiovascular system of conscious Beagle dogs. Methods Six Beagle dogs which had been successfully implanted telemetry transmitters (TL11M3-D70-PCTR) were selected as the study subjects. Four groups were designed in this study: solvent control group (0.5%CMC-Na), hawthorn fine powder low (0.24 g·kg-1), middle (0.72 g·kg-1) and high (2.16 g·kg-1) dose groups. The Beagle dogs were intragastrical administered in the order of the solvent control group, the hawthorn fine powder low-, medium-, and high-dose groups. A 7-day washout period was set between two treatments. The DSI physiological signal telemetry system was used to continuously measure the ECG, blood pressure, and body temperature from 2 h before administration to 24 h after administration. Results Compared with the solvent control group at the same time, the high-dose group had lower heart rate at 6 h and 8 h after administration, and the difference was statistically significant (P<0.05). At 6 h after administration, the QT interval of the animals in the high-dose group was prolonged. The difference was statistically significant (P<0.05), but there was no biological significance because the difference was dose-independent. Compared with the solvent control group, hawthorn fine powder had no effect on systolic blood pressure, diastolic blood pressure, mean pressure and body temperature of the experimental groups at each time points. Conclusion No significant changes were observed in the cardiovascular system of the Beagle dogs which was intragastrical administrated with hawthorn fine powder.

The male and female rats (male/female ratio was 1:2) mated since 16:00 of 28th day in all 4 groups. The vaginal smear was used to confirm pregnancy, and irrigation was stopped on the 4th day after the pregnancy. The rats were sacrificed at 16:00 on the same day,and the flow cytometry was used to detect the Thl7 and Treg cells in the peripheral blood . The expression of RORγt and Foxp3 in the endometrium was detected by immunohistochemistry. Results ①Compared with the normal control group, the expression of Foxp3 was significantly decreased in the stress group (P<0.05). There was no significant difference between the stress + Jieyuyubao group and the normal control group (P>0.05). ②Compared with the normal control group, the expression of RORγt in the endometrium of the stress group was significantly increased (P<0.05). There was no significant difference between the stress + Jieyuyubao group and the normal control group (P>0.05). ③Compared with the ratio of Th17 cell in the stress group, the ratio in the normal control group,Stress + prednisone group, and the stress + Jieyuyubao group was significantly lower (P<0.05). In addition, the ratio of Treg cells in the stress + prednisone group and the stress + Jieyuyubao group increased significantly comparing to the stress group (P<0.05). There was no significant difference in Th17/Treg between the control group and the stress + Jieyuyubao group (P>0.05). Compared with the stress group, the Th17/Treg of the remaining three groups was significantly different (P<0.05).④Th17/Treg ratio was positively correlated with RORγt expression (r=0.608,P<0.05),and Th17/Treg ratio was negatively correlated with T Foxp3 expression (r=-0.389,P<0.05). Conclusion Jieyuyubao recipe can effectively reduce the ratio of Th17/Treg in peripheral blood of rats with chronic stress, reduce the inflammatory damage, and increase the anti-stress ability of the body. High dose of Jieyuyubao recipe can significantly increase Foxp3 and reduce RORγt of endometrium in implantation window. We speculate that Jieyuyubao improves the implantation of the endometrial inflammatory response by balancing the imbalance of Th17/Treg caused by stress, thereby improving the implantation microenvironment and increasing the pregnancy rate.

Objective To establish a method to determine the water in antibiotics by headspace capillary gas chromatography-thermal conductivity detector (HCGC-TCD) method. Methods An HCGC-TCD method was established by using Agilent HP-PLOT/Q (30 m×0.530 mm,40 μm) capillary column. The high purity helium was used as the carrier gas with the flow rate of 3 mL·min-1. The injection volume was 250 μL with the split ratio of 20:1. The injection temperature was 200 ℃. The column temperature program was set as followed: initial temperature 150 ℃,keep for 6 min;then rise the temperature to 250 ℃ at a rate of 60 ℃·min-1,keep for 5 min. The detector temperature of thermal conductivity detector (TCD) was 250 ℃ and the current was 70 mA. Results The calibration curve showed a good linearity within the range of 1.717-16.667 mg·mL-1 (r=0.999 8). The average recovery was 92.76%-103.55%,with repeatability precision RSD was 4.7% (n=6). Conclusion This method is simple, quick, and accurate. The method can be used for the determination of water in the antibiotics.

Objective To improve the quality standard for Naoliqing tablets. Methods The microscopic identification of Achyranthis Bidentatae Radix,Margaritifera Concha and Pinelliae Rhizoma Praeparatum cum Aluminem were adopted. Thephysical and chemical identification was used for the identification of Haematitum and Magnetitum in Naoliqing tablets. The qualitative analysis of Achyranthis Bidentatae Radix,Fel Suillus and Borneolum Syntheticumin Naoliqing tablets were performed by thin layer chromatography (TLC). The iron element in Naoliqing tablets was determined by atomic absorption spectrophotometry (AAS). The content determination of borneol andmenthol in the drug were conducted by gas chromatography (GC). Results The microscopic identification method could be performed simply with good specificity. The physical and chemical identification had strong specificity. TLC spots were clear and well separated. GC analysis indicated that the calibration curves were linear in the ranges of 0.059 8 to 0.997 0 mg·mL-1 and 0.061 6 to 1.028 0 mg·mL-1 for menthol and borneol respectively. The average recoveries of menthol and borneol were 102.70% and 101.40% with RSDs of 1.26% and 0.92%,respectively. The results of AAS analysis showed that there was a good linear relationship of iron within the range of 0.5-10.0 mg·mL-1,the recovery of iron was 100.83% with RSD of 1.46%. Conclusion The established methods are highly specific, stable and reproducible which can be used in the evaluation and control of the quality of Naoliqing tablets.
