Objective To study the effects of Biyun spray on allergic rhinitis (AR) induced by 2,4-tolunen diisocyanate (TDI) in guinea pigs. Methods TDI was selected as sensibiligen and administered intranasally to develop the AR model.The experimental guinea pigs were randomly divided into 4 groups: control group, model control group, Biyun spray group, positive control (ketotifen fumarate) group.Corresponding volume of 0.9% sodium chloride solution was administered to the control and model control groups.In the Biyun spray group, Biyun spray was administered at a dose of 96 mg.kg-1, which was twice the clinical equivalent dose.Ketotifen fumarate was administered to positive control group at a dose of 0.07 mg.kg-1.Symptoms were observed and the nasal symptom score of AR was determined.The pathological changes of nasal mucosa tissue were observed under a microscope.Eosinophilic granulocytes (EOS) and mast cells (MC) were counted.Results Among the control, model control, Biyun spray, and positive control groups, the nasal symptom scores of AR were decreased from (1.17±0.63), (7.05±1.24), (7.56±1.05), (7.69±0.96) to (1.11±0.54), (7.33±0.67), (4.92±0.96), (5.14±0.77), respectively after the treatments.Biyun spray significantly decreased the nasal symptom score of AR (P<0.01).Pathological changes of nasal mucosa tissue were improved obviously.Among the control, model control, Biyun spray, and positive control groups, the EOS number was (1.68±0.85), (9.13±1.68), (4.51±2.25) and (3.59±1.77), respectively; the MC number was (2.23±1.26), (21.03±5.03), (14.05±6.88) and (13.14±5.41), respectively; the degranulation MC number was (1.32±1.03), (13.64±5.47), (8.49±2.37) and (6.06±2.55), respectively.Biyun spray significantly improved the pathological changes of nasal mucosa with EOSs and MCs being decreased significantly (P<0.01).Conclusion Biyun spray is effective in treating allergic rhinitis in guinea pigs.
